摘要
为筛选出具有产纤维素酶能力的菌株,以东帕米尔高原金草滩植物根际土壤为样本,采用刚果红水解圈法初步筛选到了5株菌株,分别命名为:XY-14、XY-38、XY-39、XY-49和XY-65。这5株菌株在CMC-Na培养基上长势良好,菌落均呈现圆形、并产生白色菌丝,最适生长温度为25℃。通过水解圈和滤纸崩解试验,确定XY-38、XY-39、XY-49这3株菌的酶活力较强。经分子鉴定,确定XY-38、XY-39为睾酮丛毛单孢菌(Comamonas testosteroni strain),XY-49为假单孢菌(Pseudomonas sp.strain)。菌株的生长曲线和产酶特性表明,XY-39在第6天时酶活力最大,为9.33 U/mL,XY-49在第5天时酶活力最大,为10.49 U/mL。
In order to screen for strains with cellulase production ability,this study used the rhizosphere soil of plants in the Golden Grassland of the East Pamir Plateau as a sample and preliminarily screened 5 strains using the Congo Red Hydrolysis Circle method,named XY-14,XY-38,XY-39,XY-49,and XY-65.The five strains grew well on CMC-Na medium,and the colonies were round and produced white hyphae.The optimum growth temperature was 25℃.Through the hydrolysis circle and filter paper disintegration test,it was determined that the enzyme activities of XY-38,XY-39 and XY-49 were strong.Molecular identification showed that XY-38 and XY-39 were Comamonas testosteroni strain,and XY-49 was Pseudomonas sp.strain.The growth curve and enzyme production characteristics of the strain showed that the enzyme activity of XY-39 was the highest on the 6 th day,which was 9.33 U/mL,and the enzyme activity of XY-49 was the highest on the 5 th day,which was 10.49 U/mL.
