白藜芦醇对多囊卵巢综合征小鼠心脏功能结构的改善作用及其机制

Ameliorative effect and mechanism of resveratrol on cardiac structure and function in mice with polycystic ovarian syndrome

摘要 目的观察白藜芦醇(Resveratrol,Res)对多囊卵巢综合征(polycystic ovarian syndrome,PCOS)小鼠心脏功能结构的改善作用,探讨其可能作用机制。方法取30只3周龄雌性SPF级小鼠,均分为模型组、干预组及空白组:模型组和干预组小鼠皮下注射脱氢表雄酮(6 mg/100 g)1次/天,连续21 d构建PCOS模型,空白组小鼠皮下注射6 mg/100 g芝麻油(1次/天,连续21 d)。干预组小鼠用100 mg/kg的Res灌胃,1次/天,连续28 d,模型组及空白组小鼠正常饲养。采用小动物心电图仪和超声心动图检测各组小鼠心率、QT间期和左心室射血分数;处死各组小鼠,取心脏组织称重并留取主动脉血液,采用HE染色法观察小鼠心肌组织肥大情况、Masson染色法观察小鼠心肌组织纤维化情况,采用Western Blotting法检测小鼠心肌组织M1型巨噬细胞标志物CD86、M2型巨噬细胞标志物CD206,采用ELISA法检测小鼠主动脉血清炎症因子白细胞介素6(IL-6)、精氨酸酶1(ARG-1)。结果与空白组相比,末次灌胃结束后模型组小鼠心率低、QT间期长、左心室射血分数低、心脏质量大(P<0.05)。与模型组相比,末次灌胃结束后干预组小鼠心率高、QT间期缩短、射血分数升高、心脏质量低(P<0.05)。与空白组相比,模型组小鼠心脏细胞横截面较大,纤维组织多;与模型组相比,干预组小鼠心脏细胞横截面变小,纤维组织减少。与模型组相比,空白、干预组小鼠心肌组织CD206蛋白相对表达量高、CD86蛋白相对表达量低(P均<0.05)。与空白组相比,模型组小鼠主动脉血清IL-6水平升高、ARG-1水平下降(P均<0.05);与模型组相比,干预组小鼠小鼠主动脉血清IL-6水平降低、ARG-1水平升高(P均<0.05)。结论Res灌胃可改善PCOS小鼠的心脏结构和功能、促进心肌组织CD206蛋白表达、抑制心肌组织CD86蛋白表达,降低主动脉血清IL-6水平、提高主动脉血清ARG-1水平,其机制可能为Res促进心脏巨噬细胞向M2型极化,� Objective To investigate the ameliorative effect of resveratrol(Res)on the cardiac function and struc⁃ture of mice with polycystic ovarian syndrome(PCOS)and to explore its possible mechanism of action.Methods Thirty 3-week-old female SPF mice were divided into the model group,intervention group,and blank group.Mice in the model group and the intervention group were injected subcutaneously with dehydroepiandrosterone(6 mg/100 g)once a day for 21 consecutive days to construct the PCOS models,and the mice in the blank group were injected with 6 mg/100 g sesame oil subcutaneously(once a day,for 21 days).The mice in the intervention group were gavaged with 100 mg/kg Res once a day for 28 days,and the mice in the model group and the blank group were fed normally.Animal electrocardiographs and echocardiography were used to detect heart rate,QT interval and ejection fraction in each group.The mice in each group were sacrificed,the heart tissue was weighed and the aortic blood was retained,the hypertrophy of myocardial tissue was observed by HE staining,the fibrosis of myocardial tissue was observed by Masson staining,the M1 macrophage marker CD86 and M2 macrophage marker CD206 were detected by Western blotting,and the serum inflammatory cytokines inter⁃leukin 6(IL-6)and arginase 1(ARG-1)were detected by ELISA.Results Compared with the blank group,the model group had a lower heart rate,longer QT interval,lower ejection fraction and higher heart mass after the last gavage(all P<0.05).Compared with the model group,the intervention group had a higher heart rate,shorter QT interval,increased ejection fraction and lower heart mass after the last gavage(all P<0.05).Compared with the blank group,the cross-sec⁃tional area of heart cells in mice of the model group was larger and there were more fibrous tissues;compared with the mod⁃el group,the cross-sectional area of heart cells in the intervention group became smaller and the fibrous tissue decreased.Compared with the model group,the relative expression levels of CD206 p
