秦皮水煎液干预类风湿性关节炎的作用研究

摘要 目的:观察秦皮水提取物对类风湿关节炎(RA)大鼠关节肿胀情况及组织形态、血清TNF-α、IL-1β和RF水平的影响。方法:50只雄性SD大鼠,依据随机数字表按体质量随机分为正常组、模型组、双氯芬酸钠组[3 mg/(kg·bw)]、秦皮高剂量组[1.4 g/(kg·bw)]和秦皮低剂量组[0.7 g/(kg·bw)]。以牛Ⅱ型胶原蛋白诱导建立RA大鼠模型,正常组、模型组灌胃15 mL/(kg·bw)蒸馏水;双氯芬酸钠组、秦皮高剂量组、秦皮低剂量组分别灌胃给予对应的受试药物。分别在实验第28、35、42天测量大鼠关节直径,第42天实验结束后处死大鼠,腹主动脉取血,离心取血清,酶联免疫吸附法(ELISA)测定血清中TNF-α、IL-1β、RF水平,取踝关节组织HE染色观察病理变化。结果:与正常组比较,模型组大鼠第35、42天关节直径水平显著升高(P <0.01),血清TNF-α、IL-1β、RF水平显著升高(P <0.01,P <0.05),炎性细胞浸润,骨膜增厚;与模型组比较,双氯芬酸钠组大鼠第42天关节直径水平显著降低,TNF-α、RF水平显著降低(P <0.05,P <0.01),踝关节炎性浸润改善,秦皮高剂量组大鼠第35天关节直径水平显著降低(P <0.01),血清IL-1β水平显著降低(P <0.01),踝关节炎性浸润缓解,秦皮低剂量组第35、42天关节直径水平显著降低(P <0.05),血清TNF-α、IL-1β、RF水平显著降低(P <0.05),踝关节炎性浸润减轻。结论:秦皮水煎液可改善大鼠类风湿关节炎,其作用可能与降低TNF-α、IL-1β、RF水平,改善踝关节病理组织形态有关。 Objective:To observe the effects of water extract of Cortex Fraxini(CF)on joint swelling,tissue morphology and serum levels of TNF-α,IL-1βand RF in rats with rheumatoid arthritis.Methods:According to body weight,50 male SD rats were randomly divided into normal group,model group,diclofenac sodium group(3 mg/kg·bw),CF high-dose and low-dose groups(1.4 g/kg·bw,0.7 g/kg·bw).A rat model of RA was established by induction of bovine typeⅡcollagen.Normal group and model group were gavaged with 15 mL/kg·bw distilled water,and diclofenac sodium group and CF high dose and low dose groups were gavaged with corresponding medicine respectively.Joint diameter of rats was measured on day 28,35 and 42,and the rats were euthanized after the administration on day 42.Blood was collected from abdominal aorta and serum was collected by centrifugation.The levels of TNF-α,IL-1βand RF in serum were determined by enzyme-linked immunosorbent assay(ELISA).The pathological changes were observed by HE staining of ankle joint tissues.Results:Compared with normal group,the joint diameter of rats on day 35 and 42 of model groups was significantly increased(P<0.01),the serum levels of TNF-α,IL-1βand RF were significantly increased(P<0.01 or P<0.05),and inflammatory cells were infiltrated and periosteum thickened.Compared with model group,the joint diameter of rats on day 42 of diclofenac sodium group significantly decreased,the levels of TNF-αand RF significantly decreased(P<0.05,P<0.01),and the inflammatory infiltration of ankle was improved.The joint diameter level on day 35 of CF high-dose group significantly decreased(P<0.01),serum IL-1βlevel significantly decreased(P<0.01),and ankle inflammatory infiltration relieved.Joint diameter level was significantly decreased(P<0.05),serum TNF-α,IL-1βand RF levels were significantly decreased(P<0.05)in CF low-dose group on day 35 and 42(P<0.05),and ankle inflammatory infiltration was relieved.Conclusion:The Cortex Fraxini decoction can improve rheumatoid arthritis in rats,and its effec
