摘要
目的探讨秦皮提取物及秦皮甲素对大鼠类风湿关节炎的保护作用,比较秦皮提取物与秦皮甲素的抗炎效果。方法分别采用角叉菜胶和弗氏完全佐剂制备大鼠急性关节炎和类风湿关节炎模型,并给予秦皮提取物和秦皮甲素干预后,测量大鼠足厚度、体重变化,并记录关节评分,酶联免疫吸附试验(ELISA)法测定大鼠血浆白细胞介素1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)水平及超氧化物歧化酶(SOD)活性变化。结果与空白组比较,2种炎症模型大鼠模型组的IL-1β,TNF-α,MDA水平显著上升,SOD活性显著下降(P<0.05);与模型组比较,在2种炎症模型中,秦皮提取物组和秦皮甲素组IL-1β,TNF-α含量均显著降低(P<0.05);在角叉菜胶模型中,秦皮甲素在致炎1,2 h表现出抑制作用(P<0.05),秦皮提取物对SOD活力有显著影响(P<0.05),秦皮甲素组在降低大鼠TNF-α水平上较秦皮提取物组显著(P<0.05);在佐剂模型中,秦皮提取物组和秦皮甲素组MDA含量显著降低(P<0.05),秦皮提取物组关节评分较低(P<0.05),秦皮甲素组在降低IL-1β水平上较秦皮提取物组更显著(P<0.05)。结论秦皮提取物及秦皮甲素对类风湿关节炎有一定的保护作用,秦皮甲素可能为秦皮的主要抗炎物质,其作用机制可能与调节IL-1β和TNF-α水平有关。
Objective To investigate the effect of fraxini cortex extract and aesculin on rheumatoid arthritis of rats and compare the an- ti- inflammatory effect of fraxini cortex extract and aesculin. Methods Two model groups of arthritis rats induced by adjuvant which were similar to arthritis rheumatic and acute rats induced by carrageenan which were similar to acute arthritis were made and treated with fraxini cortex extract. The paw thickness,weight changes were measured,and the joint score were recorded;the changes of IL-1β, TNF- α, MDA, and SOD in serum were detected by ELISA. Results Compared with the blank group, the levels of IL- 1β ,TNF- α,and MDA in the two model groups were significantly increased, and the activity of SOD significantly reduced (P 〈 0. 05);the contents of IL- 1β and TNF- α fraxini in the cortex extract and aesculin group were decreased significantly (P 〈 0. 05);the aesculin showed in- hibition effect on proinflammatory after 1,2 h(P 〈 0. 05),fraxini cortex extract showed significant effect on SOD(P 〈 0. 05),and aes- culin showed better effect on reducing the level of TNF-α than fraxini cortex extract in the carageen model( P 〈 0. 05). In the adju- vant model,the MDA content decreased significantly in both groups(P 〈 0. 05),the aesculin group had lower joint score(P 〈 0. 05), and aesculin showed better effect on reducing the level of IL- 1β than fraxini cortex extract( P 〈 0. 05). Conclusion Fraxini cortex extract and aesculin has certain therapeutic effect on rheumatoid arthritis, and aesculin may be a major anti- inflammatory. Its mecha- nism may be associated with IL- 1β and TNF- α levels.
