摘要
目的探讨清胃散治疗小鼠胃热型牙周炎的作用机制。方法将小鼠随机分成正常组、模型组和治疗组,连续ig辣椒醇提液,建立小鼠胃热型牙周炎模型,ig给予小鼠清胃散进行治疗。显微镜下观察小鼠胃和牙周组织的病理变化;采用酶联免疫吸附法测定各组小鼠血清中的炎症因子、氧化应激因子和凋亡因子的含量;并通过干式试剂片技术测定血清中的17种生化指标。结果模型小鼠出现了大便干燥、小便亮黄、腹胀等胃热证症状,病理切片显示胃和牙周组织有大量炎性细胞浸润,符合胃热型牙周炎的诊断标准。清胃散能显著减少胃热型牙周炎小鼠的炎性细胞,恢复胃和牙周组织结构,减少牙周充血;显著升高血清中白细胞介素-10(IL-10)、超氧化歧化酶、B细胞淋巴瘤因子2的含量,显著降低IL-1β、IL-2和IL-6的含量;显著升高尿素、肌酐、总蛋白、白蛋白和碱性磷酸酶的含量,显著降低总胆红素的含量。结论清胃散治疗胃热型牙周炎的作用机制可能与抗炎、抗氧化应激、抗凋亡、调节代谢和提升免疫有关。
OBJECTIVE To explore the therapeutic mechanism of Qingwei powder on gastric heat type periodontitis mice.METHODS The mice were randomly divided into normal group,model group and treatment group.The gastric heat periodontitis model of mice was established by continuous intragastric administration of capsicum alcohol extract,and the model was treated by three doses of Qingwei powder.The pathological changes of gastric and periodontal tissues were observed and recorded under microscope.The contents of inflammatory cytokines,oxidative stress factors and apoptosis factors in serum were determined by ELISA.The changes of 17 biochemical indexes in serum were investigated by dry reagent tablet technique.RESULTS The model mice showed symptoms of gastric heat such as dry stool,bright yellow urine and abdominal distension.Pathological sections showed a large number of inflammatory cells infiltrating the stomach and periodontal tissue,which met the diagnostic criteria of gastric heat type periodontitis.Qingwei powder significantly reduced inflammatory cells,restored gastric and periodontal tissue structure,and reduced periodontal congestion in mice with gastric heat type periodontitis.The levels of IL-10,SOD and Bcl-2 in serum were significantly upregulated,and the levels of IL-β,IL-2 and IL-6 were significantly downregulated.The levels of urea,creatinine,total protein,albumin and alkaline phosphatase were significantly increased,while the levels of total bilirubin were significantly decreased.CONCLUSION The mechanism of Qingwei powder in the treatment of gastric heat type periodontitis may be related to anti-inflammation,anti-oxidative stress,anti-apoptosis,metabolism regulation and enhancement of immunity.
