摘要
以环孢素A为模型药,选用卡波姆和羟丙甲纤维素为凝胶基质制备环孢素A眼用pH敏感型凝胶,选择胶凝能力和流动性为评价指标,通过正交实验筛选最优处方,并对其进行质量评价。结果表明,最佳凝胶处方基质为0.2%卡波姆和1.5%的羟丙甲纤维素,制备的凝胶在72 h内的药物累积释放量及累积溶蚀量均达到了70%以上,具有较好的缓释作用。因此,本文制备的凝胶可以满足眼部用药要求,可以有效延长药物在眼部的滞留时间。
Cyclosporine A as a model drug was selected.Carbopol and hydroxypropyl methylcellulose as thermosensitive materials were used to design and prepare pH-sensitive gel for ocular administration.The concentration of carbopol and hydroxypropyl methylcellulose was evaluated with index of mobility and gelling ability by orthogonal experiment.The quality of gel was evaluated includes with morphology and release.The results showed that the optimal prescription of formulation were carbopol and hydroxypropyl methylcellulose were 0.2%and 1.5%.The cumulative drug release and dissolution of the prepared gel reached more than 70%within 72 hours,which had a good sustained-release effect.Therefore,the gel prepared in this paper can meet the requirements of eye medication and extend the retention time of drugs in the eye effectively.
