摘要
目的:探讨甘草次酸金配合物(GA-Au)通过抑制高迁移率族蛋白B1(HMGB1)的核质易位调控自噬抑制肌腱干细胞(TSCs)氧化应激发挥抗肌腱病的作用。方法:MTT法筛选造模及给药浓度;Western Blot检测HMGB1、P62、BECN1的蛋白表达;还原型谷胱甘肽(GSH)和丙二醛(MDA)试剂盒检测细胞内GSH、MDA水平;免疫荧光检测HMGB1的表达情况;构建HMGB1过表达质粒,验证GA-Au的作用;HE和Masson染色观察大鼠肌腱组织胶原排列。结果:与造模组比较,GA-Au给药缓解了氧化应激状态下,TSCs内GSH的耗竭和MDA的积聚。体内实验表明,给药组的大鼠肌腱组织较造模组胶原排列较为紧密,未出现明显的空泡化以及肌腱断裂的情况。结论:GA-Au通过抑制HMGB1核质易位调控自噬,抑制TSCs氧化应激从而发挥抗肌腱病作用。
Objective:To investigate the alleviation of glycyrrhetinic acid-gold complex(GA-Au)on tendinopathy by inhibiting oxidative stress of tendon stem cells(TSCs)via nucleoplasmic translocation of high mobility group protein B1(HMGB1).Methods:MTT was used to screen the concentration of molding and drug administration.The protein expression of HMGB1,P62 and BECN1 were detected by Western Blot.The levels of intracellular reduced glutathione(GSH)and malondialdehyde(MDA)were detected by the corresponding kits.The expressions of HMGB1 wsa detected by immunofluorescence.HMGB1overexpression plasmid was constructed to verify the effect of GA-Au.The collagen arrangement of rat tendon was observed by HE and Masson staining.Results:Compared with the modeling group,GA-Au administration alleviated the depletion of GSH,the accumulation of MDA in TSCs under oxidative stress.In vivo,the collagen arrangement of the tendon tissue in the GA-Au group was more collected than that in the modeling group,and there was no obvious vacuolation and tendon rupture.Conclusion:GA-Au can inhibit TSCs oxidative stress by regulating HMGB1 nucleoplasmic translocation and autophagy,thus playing an antitendinopathy role.
