摘要
目的:优选壮药龙路通颗粒提取工艺。方法:用高效液相色谱法测定三七皂苷R_(1),人参皂苷Rg_(1)和Rb_(1),并以出膏率、三七皂苷R_(1),人参皂苷Rg_(1)和Rb_(1)含量为评价指标。采用单因素法考察加水量、提取时间和提取次数,并通过正交试验优选壮药龙路通颗粒最佳提取工艺。结果:以三七皂苷R_(1),人参皂苷Rg_(1)和Rb_(1)的含量作为龙路通颗粒质控标准,测得龙路通颗粒中3种指标性成分的质量分数分别为三七皂苷R_(1)45.6~60.6 mg/g,人参皂苷Rg_(1)159.3~234.4 mg/g,和人参皂苷Rb_(1)143.8~246.0 mg/g;龙路通颗粒最佳提取工艺为加水煎煮提取3次,第一次加12倍量水,第二、三次分别加10倍量水,每次1.5 h。结论:优选的提取工艺合理可行,为壮药龙路通颗粒的生产及进一步开发提供了实验依据。
Objective:To optimize the extraction process of Zhuang medicine Longlutong granules.Method:The content determination method of Notoginsenoside R_(1),Ginsenoside Rg_(1),Ginsenoside Rb_(1)was established by HPLC.The extraction amount,Notoginsenoside R_(1)content,Ginsenoside Rg_(1)content,Ginsenoside Rb_(1)content were used as evaluation indexes.The single factor method was used to investigate the amount of water added,extraction time,and extraction frequency,and the optimal extraction process for Zhuangyao Longlutong decoction was optimized through orthogonal test.Result:Taking the contents of Notoginsenoside R_(1),Ginsenoside Rg_(1),Ginsenoside Rb_(1)as the quality control standard of Longlutong granules,it was determined that the mass fractions of the three index components in Longlutong granules were Notoginsenoside R_(1)45.6~60.6 mg/g,Ginsenoside Rg_(1)159.3~234.4 mg/g,and Ginsenoside Rb_(1)143.8~246.0 mg/g;The optimal extraction process is to decoct with water three times,adding 12 times the amount of water for the first time,and adding 10 times the amount of water for the second and third times,each time for 1.5 hours.Conclusion:The optimized extraction process is reasonable and feasible,providing experimental basis for the production and further development of Zhuang meidicine Longlutong granules.
