基于PI3K/Akt信号通路探讨胃复春对胃癌前病变大鼠的干预作用

Intervention Effect of Weifuchun on Precancerous Lesions of Gastric Cancer Rats Based on PI3K/Akt Signaling Pathway

摘要 目的 观察胃复春对胃癌前病变(Precancerous lesions of gastric cancer,PLGC)大鼠的干预作用。方法 选取健康SPF级雄性Wistar大鼠30只,按随机数字表法分为胃复春组、模型组、对照组,每组各10只,胃复春组给予胃复春灌胃,模型组及对照组给予生理盐水灌胃,1次/d,共灌胃16周。造模成功后,立即处死所有大鼠,按照组别取胃黏膜组织,进行病理组织学观察。其余组织应用RT-PCR法、Western Blot法检测Akt、p-Akt、NF-κB、COX-2的表达情况。结果 胃复春可以有效的改善PLGC大鼠模型的一般状态、宏观表征等,还可逆转大鼠胃黏膜的病理学状态,从而降低PLGC的发生率;与对照组比较,模型组大鼠胃黏膜组织Akt、p-Akt、NF-κB、COX-2蛋白表达水平均明显升高,差异有统计学意义(P<0.05);与模型组比较,胃复春组Akt、p-Akt、NF-κB、COX-2蛋白水平均明显降低,差异有统计学意义(P<0.05)。与对照组比较,模型组大鼠胃黏膜组织Akt、p-Akt、NF-κB、COX-2 mRNA表达水平均明显升高,差异有统计学意义(P<0.05);与模型组比较,胃复春组大鼠胃黏膜组织Akt、p-Akt、NF-κB、COX-2 mRNA表达水平均明显降低,差异有统计学意义(P<0.05)。结论 胃复春可通过抑制Akt、p-Akt、NF-κB、COX-2增殖,调控PI3K/Akt信号通路的异常活化,进一步阻断PLGC的发生与发展。 Objective To observe the effect of Weifuchun on precancerous lesions of gastric cancer rats(PLGC).Methods Thirty healthy,SPF,male Wistar rats were selected and divided into a Weifuchun group(n=10),a model group(n=10),and a control group(n=10)by random number table.The Weifuchun group received Weifuchun via gavage,while the model and control groups were administered saline or corresponding drug treatment,once daily for 16 weeks.Following successful modeling,all rats were euthanized,and gastric mucosa tissues were obtained for histopathological analysis.The expressions of Akt,p-Akt,NF-κB,and COX-2 in tissue samples were detected using RT-PCR and Western Blot.Results Weifuchun improved the general condition and macroscopic features of the PLGC rat model,and reversed the pathological state of the gastric mucosa,thus reducing the incidence of PLGC.Compared with the conditions in the control group,the protein expressions of Akt,p-Akt,NF-κB,and COX-2 in the model group were increased(P<0.05).The Weifuchun group had decreased levels of these proteins compared with the model group(P<0.05).Similarly,the mRNA expressions of Akt,p-Akt,NF-κB,and COX-2 were elevated in the model group compared with the control group(P<0.05),while reduced in the Weifuchun group compared with the model group(P<0.05).Conclusion Weifuchun may regulate the abnormal activation of the PI3K/Akt signaling pathway by inhibiting the proliferation of Akt,p-Akt,NF-κB,and COX-2,thus preventing the occurrence and progression of PLGC.
