M1型乙酰胆碱受体在微波辐射致小鼠认知行为改变中的作用

摘要 为了研究微波辐射对海马乙酰胆碱受体(AChRs)表达的影响,探讨微波辐射敏感的AChRs亚型及其在微波辐射致认知行为改变中的作用。本试验以C57BL/6N小鼠为试验对象,建立微波辐射动物模型,通过脑组织石蜡切片和苏木精-伊红(H.E.)染色,观察海马阿蒙氏角1(CA1)、阿蒙氏角2(CA2)、阿蒙氏角3(CA3)和齿状回(DG)各亚区组织病理学变化;利用实时荧光定量PCR(RT-qPCR)检测海马组织AChRs的表达;微波辐射后,采用M1型AChR激动剂VU0357017 hydrochloride以10 mg/(kg·bw)腹腔注射小鼠,利用新物体识别和Y迷宫试验评价小鼠认知行为改变。结果显示,微波辐射后7 d,小鼠海马CA3区病理改变较为明显,主要表现为神经元核固缩深染、血管周围间隙增宽;RT-qPCR结果显示,与空白对照组相比较,微波辐射后海马组织M1和M3型AChR mRNA相对表达量均显著下调(M1-AChR,P<0.01;M3-AChR,P<0.05),而M2、M4、α4和α7型AChR mRNA相对表达量均显著上调(P<0.05);新物体识别试验结果显示,与空白对照组相比较,微波辐射后小鼠的辨别指数(DI)和认知指数(RI)均显著降低(P<0.05);而VU0357017 hydrochloride给药后,与辐射对照组相比较,辐射给药组小鼠的DI和RI明显升高(P<0.05),且与空白对照组无显著性差异(P>0.05);Y迷宫试验结果显示,辐射前后小鼠的自由交替率均未见统计学差异(P>0.05)。结果表明,微波辐射可引起海马AChRs亚基构成比发生变化,海马CA3区组织病理学改变,M1型AChR对于微波辐射所致认知行为功能下降具有重要的调控作用。 To investigate the effect of microwave radiation on the expression of hippocampal acetylcholine receptors(AChRs)and explore the microwave radiation-sensitive AChR subtypes and their roles in microwave radiation-induced cognitive behavior changes.C57BL/6N mice were used as experimental subjects to establish a microwave radiation animal model.Brain tissue paraffin sections and hematoxylin-eosin(H.E.)staining were used to observe histopathological changes in the hippocampal subregions,including Cornu ammonis 1(CA1),Cornu ammonis 2(CA2),Cornu ammonis 3(CA3),and dentate gyrus(DG).Real-time fluorescence quantitative PCR(RT-qPCR)was performed to detect the expression of AChRs in the hippocampas.After microwave radiation,mice were intraperitoneally injected with the M1-type AChR agonist VU0357017 hydrochloride at a dose of 10 mg/(kg·bw),and cognitive behavior changes were evaluated using the novel object recognition and Y-maze tests.The results showed that 7 days after microwave radiation,the pathological changes in the CA3 region of the hippocampus were more pronounced in mice,mainly manifested as neuronal nuclear condensation and deep staining,as well as widening of the interstitial space around blood vessels.RT-qPCR results showed that compared with the blank control group, the relative expression levels of M1- and M3- AChR mRNA in thehippocampas were significantly downregulated after microwave radiation (M1-AChR, P <0.01;M3-AChR, P <0.05), while the relative expression levels of M2-, M4-, α4-, and α7-AChR mRNA were significantly upregulated ( P <0.05). Novel object recognition test results showed that compared with the blank control group, the discrimination index (DI) and recognition index (RI) of mice were significantly decreased after microwave radiation ( P <0.05).
