摘要
目的评价采用干姜、肉桂灌胃结合咪喹莫特乳膏涂抹,建立血热型银屑病动物模型的皮损和体内微环境变化情况。方法48只8周龄健康BALB/c小鼠(雌雄各半)随机分为正常对照组和银屑病模型组。第1天起,模型组给予0.9 g/mL干姜、肉桂水煎液,按0.1 mL/10 g·d剂量灌胃,对照组给予生理盐水0.1 mL/10 g·d剂量灌胃,持续12 d。灌胃第6天起,模型组背部涂抹咪喹莫特乳膏42 mg,对照组背部涂抹等量凡士林乳膏,每天观察皮损情况,持续7 d。第13天,小动物麻醉机麻醉后,采集两组小鼠血液,ELISA法检测两组小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、干扰素-γ(interferon-γ,IFN-γ)、白细胞介素21(interleukin-21,IL-21)、白细胞介素17(interleukin-17,IL-17)含量变化;取两组小鼠背部涂抹处皮肤,制作病理学组织切片观察皮损情况。结果模型组小鼠涂抹部位皮肤表皮增厚,表面凹凸不平,有红斑散在分布。显微镜下显示表皮角化不全、棘层增厚、皮脂腺增多;血清中TNF-α、IFN-γ、IL-21、IL-17水平均升高,与对照组比较,差异有统计学意义(P<0.05)。结论BALB/c小鼠经干姜、肉桂水煎液灌胃12 d,期间第6天起背部加涂咪喹莫特乳膏,连续涂抹7d,成功构建的血热型银屑病动物模型,皮损变化和体内微环境改变符合银屑病患者特点。
Objective To evaluate the changes in skin lesions and in vivo microenvironment of an animal model of blood heat psoriasis using Rhizoma Zingiberis and Cinnamomum Cassia combined with imiquimod cream application.Methods 488-week-old healthy BALB/c mice(half male and half female)were randomly divided into a normal control group and a psoriasis model group.Starting from the 1st day,the model group was given 0.9 g/mL of Rhizoma Zingiberis and Cinnamomum Cassia decoction by gavage at a dose of 0.1 mL/10 g·d,while the control group was given 0.1 mL/10 g·d of physiological saline by gavage for 12 days.Starting from the 6th day of gastric lavage,the model group applied 42 mg of imiquimod cream on the back,while the control group applied an equal amount of vaseline cream on the back.The skin lesions were observed daily for 7 days.On the 13th day,after anesthesia with a small animal anesthesia machine,blood was collected from two groups of mice,and ELISA was used to detect the levels of serum in both groups of tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),interleukin-21(IL-21)and interleukin-17(IL-17).Skin samples were taken from the back of two groups of mice for pathological tissue sectioning to observe the condition of skin lesions.Results The skin epidermis at the application site of the model group mice thickened,with uneven surface and scattered erythema.Under the microscope,it showed incomplete keratinization of the epidermis,thickening of the spinous layer,and increased sebaceous glands;The levels of TNF-α,IFN-γ,IL-21 and IL-17 in serum both increased,and compared with the control group,the difference was statistically significant(P<0.05).Conclusion BALB/c mice were orally administered a decoction of Rhizoma Zingiberis and Cinnamomum Cassia for 12 days,and from the 6th day onwards,imiquimod cream was added to the back for 7 consecutive days.A blood heat psoriasis animal model was successfully constructed,and the changes in skin lesions and internal microenvironment were consistent with the characterist
