摘要
目的探讨长链非编码RNA(lncRNA) POU3F3在黑色素瘤中的表达变化, 以及lncRNA POU3F3与黑色素瘤转移的关系。方法选取来郑州大学第一附属医院2022年1月至2023年5月收治并接受手术治疗的47例患者的临床样本作为研究对象, 采用荧光定量聚合酶链反应(PCR)技术分析黑色素瘤和癌旁组织lncRNA POU3F3的表达水平, 并采用免疫组织化学分析肿瘤组织和癌旁组织中黏着斑激酶(FAK)、β-连环蛋白(β-catenin)、N-钙黏蛋白(N-Cadherin)和锌指转录因子Snail蛋白的表达水平。相关性分析lncRNA POU3F3和转移相关蛋白水平的关系。采用Transwell分析lncRNA POU3F3敲降对细胞迁移和侵袭的影响。组间计量数据比较采用t检验。相关性分析采用皮尔森分析。结果癌旁组织中lncRNA POU3F3表达水平(1.23±0.18)明显低于黑色素瘤组织表达水平(2.31±0.19), 差异有统计学意义(t=27.890, P<0.05)。癌旁组织中FAK和β-catenin表达水平(58.98±11.11、73.38±12.26)明显低于黑色素瘤组织(96.28±11.74、129.60±14.61), 差异有统计学意义(t=16.660、20.200, P<0.05)。癌旁组织中N-Cadherin和Snail表达水平(45.83±13.49、55.47±7.35)明显低于黑色素瘤组织(82.17±21.99、93.47±11.70), 差异有统计学意义(t=8.846、18.860, P<0.05)。lncRNA POU3F3表达水平与FAK、β-catenin、N-Cadherin和Snail表达水平呈正相关(r=0.587、0.671、0.814、0.344, P<0.05)。lncRNA POU3F3敲降细胞迁移和侵袭数量[(45.75±9.19)、(36.13±8.76)个]明显低于对照组细胞[(92.50±5.04)、(66.13±9.51)个], 差异有统计学意义(t=12.610, 6.564, P<0.05)。结论 lncRNA POU3F3在黑色素瘤组织中表达显著上调, 并与肿瘤组织中转移相关蛋白FAK、β-catenin、N-Cadherin和Snail表达呈正相关。敲降lncRNA POU3F3可显著抑制黑色素瘤细胞的转移特性。
Objective To explore the expression changes of long non-coding RNA(lncRNA)POU3F3 in melanoma and the relationship between lncRNA POU3F3 and melanoma metastasis.Methods Clinical samples from 47 patients admitted to our hospital from January 2022 to May 2023 who underwent surgical treatment were selected as the research subjects.The expression levels of lncRNA POU3F3 in melanoma and adjacent tissues was analyzed by fluorescence quantitative polymerase chain reaction(PCR).The expression of focal adhesion kinase(FAK),β-catenin,N-cadherin,and zinc finger transcription factor Snail protein was analyzed by immunohistochemistry in tumor and adjacent tissues.The relationship between lncRNA POU3F3 and transfer related proteins(FAK,β-catenin,N-cadherin,and zinc finger transcription factor Snail protein)was analyzed.The effects of lncRNA POU3F3 knockdown on cell migration and invasion was analyzed by Transwell.The comparison of inter group measurement data was done by the t-test.Pearson analysis was used for correlation analysis.Results The expression level of lncRNA POU3F3 in adjacent cancerous tissues(1.23±0.18)was significantly lower than that in melanoma tissues(2.31±0.19,t=27.890,P<0.05).The expression level of FAK andβ-catenin(58.98±11.11,73.38±12.26)was significantly lower than that of melanoma tissue(96.28±11.74,129.60±14.61,t=16.660,20.200,P<0.05).The expression levels of N-Cadherin and Snail in adjacent cancerous tissues(45.83±13.49,55.47±7.35)were significantly lower than those in melanoma tissues(82.17±21.99,93.47±11.70,t=8.846,18.860,P<0.05).The expression level of lncRNA POU3F3 and its correlation with the expression levels of FAK,β-catenin,N-Cadherin,and Snail were positively correlated(r=0.587,0.671,0.814,0.344,P<0.05).The number of migrative and invasive cells with lncRNA POU3F3 knockdown[(45.75±9.19),(36.13±8.76)cells]was significantly less than that in the control group[(92.50±5.04),(66.13±9.51)cells,t=12.610,6.564,P<0.05].Conclusion lncRNA POU3F3 is significantly upregulated in melanoma
