摘要
目的探讨喉癌组织中长链非编码RNA(lncRNA)LUCAT1的表达水平,及其与患者临床病理特征、预后和新辅助化疗疗效的关系。方法选取2020年12月到2022年12月商丘市第一人民医院手术切除的73例喉癌标本作为研究对象,取周围的癌旁组织作为对照样本。采用荧光定量聚合酶链反应(PCR)分析肿瘤组织和癌旁组织中lncRNA LUCAT1表达水平;分析lncRNA LUCAT1与喉癌患者临床病理特征的关系;分析影响喉癌的预后因素;分析lncRNA LUCAT1与新辅助化疗疗效的关系。组间计量数据比较采用t检验。结果癌旁组织中lncRNA LUCAT1表达水平(1.10±0.15)明显低于喉癌组织中表达水平(2.18±0.32),差异有统计学意义(t=26.510,P<0.05)。Ⅲ~Ⅳ期患者肿瘤组织lncRNA LUCAT1表达水平(2.39±0.24)明显高于Ⅰ~Ⅱ期患者(1.99±0.24),差异有统计学意义(t=7.045,P>0.05)。低分化患者肿瘤组织lncRNA LUCAT1表达水平(2.39±0.25)明显高于中高分化患者(1.98±0.23),差异有统计学意义(t=6.760,P>0.05)。淋巴结转移患者肿瘤组织lncRNA LUCAT1表达水平(2.46±0.16)明显高于中高分化患者(1.90±0.12),差异有统计学意义(t=16.970,P>0.05)。lncRNA LUCAT1高表达组患者3年累计生存率(51.42%,18/35)明显低于lncRNA LUCAT1低表达组患者(73.68%,28/38),差异有统计学意义(Log-rank=4.294,P<0.05)。病理学分级、RNM分期、淋巴结转移和lncRNA LUCAT1表达水平是影响喉癌预后不良的主要因素(P<0.05)。淋巴结转移和lncRNA LUCAT1表达水平是影响喉癌患者不良预后的独立危险因素(P<0.05)。lncRNA LUCAT1高表达组患者缓解率(40.00%)显著低于低表达患者(57.89%),差异有统计学意义(χ^(2)=3.109,P<0.05)。结论lncRNA LUCAT1在喉癌组织中表达上调,与患者淋巴结转移、TNM分期和分化程度密切相关,是喉癌患者预后的独立危险因素,且与喉癌术后化疗疗效明显相关。
Objective To investigate the expression level of long non-coding RNA(lncRNA)LUCAT1 in laryngeal cancer tissue and its relationship with clinicopathological characteristics,prognosis,and neoadjuvant chemotherapy efficacy of patients.Methods Totally,73 laryngeal cancer specimens surgically removed from Shangqiu First People’s Hospital from December 2020 to December 2022 were selected as the research subjects,and surrounding cancerous tissues were taken as control samples.Fluorescence quantitative polymerase chain reaction(PCR)was used to analyze the expression level of lncRNA LUCAT1 in tumor tissue and adjacent tissues.The relationship between lncRNA LUCAT1 and clinicopathological characteristics of laryngeal cancer patients was analyzed.The prognostic factors affecting laryngeal cancer was analyzed.The relationship between lncRNA LUCAT1 and the efficacy of neoadjuvant chemotherapy was analyzed.The measurement data were compared between groups using t-test.Results The expression level of lncRNA LUCAT1 in adjacent tissues(1.10±0.15)was significantly lower than that in laryngeal cancer tissues(2.18±0.32,t=26.510,P<0.05).The expression level of lncRNA LUCAT1 in tumor tissue of stageⅢ-Ⅳpatients(2.39±0.24)was significantly higher than that of stageⅠ-Ⅱpatients(1.99±0.24,t=7.045,P>0.05).The expression level of lncRNA LUCAT1 in tumor tissue of patients with low differentiation(2.39±0.25)was significantly higher than that of patients with medium to high differentiation(1.98±0.23,t=6.760,P>0.05).The expression level of lncRNA LUCAT1 in tumor tissue of patients with lymph node metastasis(2.46±0.16)was significantly higher than that of patients without lymph node metastasis(1.90±0.12,t=16.970,P>0.05).The 3-year cumulative survival rate[51.42%(18/35)]of patients with high expression of lncRNA LUCAT1 was significantly lower than that of patients with low expression of lncRNA LUCAT1[73.68%(28/38),Log rank=4.294,P<0.05].Pathological grading,RNM staging,lymph node metastasis,and lncRNA LUCAT1 expression level were
