摘要
目的:研究侧柏叶有效成分总黄酮的最佳提取工艺,并评价有效提取成分对酪氨酸酶活性的影响。方法:采用单因素试验、正交试验考察了提取过程中各因素对侧柏叶总黄酮含量的影响;采用蘑菇酪氨酸酶多巴速率氧化法考察提取成分对酪氨酸酶活性的影响。结果:本实验确定最优的提取工艺为90%乙醇作为提取液,按照1∶15的料液比(g∶mL),以70℃的超声温度进行提取,超声时间为60 min,此条件下总黄酮提取量为1.394 mg/g。0.5 g/mL乙酸乙酯提取物的激活率最小,为-211.68%±2.42%;0.5 g/mL石油醚提取物的激活率最大,为20.30%±1.32%。结论:本研究确定了侧柏叶有效成分提取的最佳工艺,经过对有效成分活性测试可知,乙醇粗提取物和乙酸乙酯提取物对酪氨酸酶的活性有抑制作用且随着浓度升高而升高,石油醚提取物低浓度有抑制作用,高浓度有激活作用。
Objective:To study the optimal extraction process of total flavonoids,an effective component of Platycladus orientalis leaves,and evaluate the effect of effective component on tyrosinase activity.Methods:Single factor experiments and orthogonal experiments are used to investigate the effects of various factors during the extraction process on the total flavonoid content of Platycladus orientalis leaves;the mushroom tyrosinase dopa rate oxidation method is used to investigate the activity of effective component on tyrosinase.Results:The optimal extraction process is as follows:90%ethanol in the extraction solution,a material to liquid ratio of 1 to 15(g∶mL),ultrasound temperature of 70℃,and ultrasound time of 60 min.The total brass extraction under these conditions is 1.394 mg/g.The activation rate of 0.5 g/mL ethyl acetate extract is the lowest,which is-211.68%±2.42%,and the activation rate of 0.5 g/mL petroleum ether extract is the highest,which is 20.30%±1.32%.Conclusions:This experiment has determined the optimal process for extracting the active ingredients from the leaves of Platycladus orientalis.After testing the activity of the active ingredients,it is found that the crude ethanol extract and ethyl acetate extract have inhibitory effects on the activity of tyrosinase and increase with increasing concentration.The petroleum ether extract has inhibitory effects at low concentrations,and activation effects at high concentrations.
