摘要
建立了青稞酒中乳酸和乙酸的高效液相色谱法(紫外检测器)测定方法。样品用水稀释后,经反相色谱分离,以磷酸水溶液(0.12%体积分数)-甲醇(49∶1,v/v)为流动相,流速为0.6 mL/min,进样量为10μL,紫外检测器检测,检测波长为214 nm。以保留时间定性,外标法定量。结果表明:乳酸线性范围为0.0192 g/L~0.48 g/L,乙酸线性范围为0.0664 g/L~1.6624 g/L,相关系数R2=0.9999,回收率为97.66%~98.75%,RSD<3%,乳酸方法检出限为0.005 mg/mL。该方法高效便捷、重现性好,为青稞酒中乳酸和乙酸含量检测提供了准确可靠的依据。
A method for the determination of lactic acid and acetic acid in Qingke BaiJiu by high performance liquid chromatography(UV detector)was established.After the sample is diluted with water,it is separated by reverse-phase chromatography,using phosphoric acid aqueous solution(0.12%volume fraction)-methanol(49:1,v/v)as the mobile phase,the flow rate is 0.6 mL/min,the injection volume is 10 uL,and the detection wavelength is 214 nm.Qualitative analysis is based on retention time and quantitative analysis is based on external standard method.The results showed that the linear range of lactic acid was 0.0192g/L~0.48g/L,the linear range of acetic acid was 0.0664g/L~1.6624g/L,the correlation coefficient R2=0.9999,the recovery rate was 97.66%~98.75%,RSD<3%,and the detection limit of lactic acid method was 0.005mg/mL.This method is efficient,convenient and reproducible,and provides an accurate and reliable basis for the determination of lactic acid and acetic acid in Qingke BaiJiu.
