摘要
目的探讨桃红四物汤治疗子宫异常出血(AUB)的潜在生物学机制。方法通过中药系统药理数据库(TCMSP)筛选TSD的活性化合物及其靶点,运用GeneCards、OMMI等数据库数据库筛选AUB靶点,确定交集基因;构建靶点网络和蛋白质相互作用(PPI)网络,采用米非司酮、米索前列醇联合复制AUB模型,对关键蛋白进行体内验证。结果筛选得到TSD活性成分69个,对应靶点224个,AUB相关靶点3271个,交集靶点164个。实验结果显示,TSD可显著减少大鼠子宫的出血量和出血时间,降低模型大鼠血清中IL-6和TNF-α,下调Akt1、TNF-α蛋白的表达。结论TSD对AUB的治疗作用可能与降低IL-6和TNF-α炎症因子的水平、下调Akt1和TNF-α蛋白表达有关,为进一步开展TSD治疗AUB的机制研究提供了新思路与方法。
Objective To investigate the potential biological mechanism of TaoHong SiWu decoction(TSD)for abnormal uterine bleeding(AUB).Methods The active compounds and their targets of TSD were screened by the Traditional Chinese Medicine Systematic Pharmacology Database(TCMSP),and the AUB targets were screened using GeneCards,OMMI and other database databases to identify intersecting genes;the target network and protein interaction(PPI)network were constructed,and the AUB model was replicated using the combination of mifepristone and misoprostol,and the key proteins were in vivo validation.Results The screening yielded 69 active components of TSD,corresponding to 224 targets,3271 AUB-related targets and 164 intersecting targets.The experimental results showed that TSD significantly reduced the bleeding volume and bleeding time in rat uterus,decreased IL-6 and TNF-α in serum of model rats,and down-regulated the expression of Akt1 and TNF-αprotein.Conclusion The therapeutic effect of TSD on AUB may be related to the reduction of IL-6 and TNF-α inflammatory factor levels and down-regulation of Akt1 and TNF-α protein expression,which provides new ideas and methods for further research on the mechanism of TSD for AUB.
