抗阻、耐力运动对肥胖小鼠肝脏自噬相关蛋白表达的影响

摘要 为探讨抗阻、耐力运动对肥胖小鼠肝脏自噬相关蛋白活性的影响,将100只4周龄C57BL/6雄性小鼠进行8周高脂饲料喂养,得到32只肥胖小鼠。将肥胖小鼠随机分为4组,分别为高脂喂养对照组(HHC组)、普通饲料对照组(HNC组)、普通饲料+有氧运动组(HNE组)、普通饲料+抗阻运动组(HNR组),每组8只,HNE和HNR组分别进行8周跑台和爬梯训练。ELISA法检测血糖及肝脏中总胆固醇(TC)和总三酰甘油(TG)含量,Western blot法检测小鼠肝脏自噬相关蛋白表达量。结果表明:8周高脂膳食干预后,小鼠肝脏自噬相关蛋白Beclin-1表达量显著降低,p62蛋白表达量显著升高;8周抗阻和耐力运动可显著降低肥胖小鼠血清和肝脏TG和TC含量,小鼠肝脏自噬相关蛋白Beclin-1表达量、LC3Ⅱ/Ⅰ比值均显著升高,p62蛋白表达量显著降低,不同运动形式间3种自噬相关蛋白均未发现显著差异。综上,高脂膳食可使肝脏自噬水平降低,体内脂质积累增加,而抗阻和耐力运动均可促进肥胖小鼠肝脏自噬水平提高,促进肝脏脂质代谢,且2种运动形式间无显著差异,抗阻和耐力运动均可作为预防和治疗肥胖及其相关肝脏疾病的辅助手段。 To explore the effects of resistance and endurance exercise on liver autophagy related protein activity in obese mice, 100 four week old C57BL/6 male mice were fed with high-fat diet for eight weeks, resulting in 32 obese mice. Obese mice were randomly divided into four groups: high-fat diet control group(HHC), normal diet control group(HNC), normal diet+aerobic exercise group(HNE), and normal diet+resistance exercise group(HNR), with 8 mice in each group.The HNE and HNR groups received eight weeks of treadmill and ladder training,respectively.ELISA method was used to measure blood glucose and TC and TG levels in the liver,while Western blot method was used to measure the expression of autophagy related proteins in the mouse liver.The results showed that after eight weeks of high-fat diet intervention,the expression of the autophagy related protein Beclin-1 in the liver of mice was significantly reduced,while the expression of p62 protein was significantly increased;Eight-week resistance and endurance exercise can reduce the levels of TG and TC in the serum and liver of obese mice.The expression of the autophagy related protein Beclin-1 and the LC3 Ⅱ/Ⅰ ratio in the liver of mice were significantly increased,while the expression of p62 protein was significantly reduced.No significant differences were found among the three autophagy related proteins among different exercise groups.To sum up,a high-fat diet can reduce liver autophagy levels and increase the accumulation of lipids in the body.Both resistance and endurance exercise can promote liver autophagy levels and liver lipid metabolism in obese mice,and there is no significant difference between the two types of exercise.Resistance and endurance exercise can be used as auxiliary means to prevent and treat obesity and related liver diseases.
