摘要
目的探索参苓白术散通过Linc01615/miR-491-5p对结直肠癌细胞凋亡、细胞周期和侵袭作用的影响。方法通过细胞转染构建分组:空白对照组、参苓白术散组、参苓白术散组+siRNA Linc01615、参苓白术散组+oe Linc01615、参苓白术散组+miR-491-5p mimics、参苓白术散组+miR-491-5p抑制剂组、参苓白术散组+miR-491-5p mimics+oe Linc01615,加入相应的血清进行干预。经相应的血清处理后,Annexin V-FITC/PI双染法流式细胞仪检测细胞凋亡、细胞周期分布情况,transwell实验检测细胞侵袭情况。结果参苓白术散处理细胞后,抑制细胞增殖、侵袭及细胞周期的G0/G1期并促进凋亡,在分别加入siRNA Linc01615和miR-491-5p mimics后效果更为显著,差异具有统计学意义(P<0.05)。结论参苓白术散可能通过Linc01615/miR-491-5p抑制LoVo细胞的侵袭,促进凋亡并将细胞周期阻滞于G0/G1期。
Objective:To study the effect of Shenling Baizhu Powder on apoptosis,cell cycle and invasion of colorectal cancer via Linc01615/miR-491-5p.Methods:Groups were constructed by cell transfection and divided blank control group,Shenling Baizhu Powder group,Shenling Baizhu Powder group+siRNA Linc01615,Shenling Baizhu Powder group+oe Linc01615,Shenling Baizhu Powder group+miR-491-5p mimics,Shenling Baizhu Powder group+miR-491-5p inhibitor group,Shenling Baizhu Powder group+miR-491-5p mimics+oe Linc01615.All groups were added to the corresponding serum for intervention.After treatment with the corresponding serum,Annexin V-FITC/PI double staining flow cytometry was used to detect apoptosis and cell cycle distribution,and transwell assay was used to detect cell invasion.Results:Shenling Baishu Powder inhibited cell proliferation,invasion and G0/G1 phase of cell cycle,and promoted apoptosis,and the effect was more significant after the addition of siRNA Linc01615 and miR-491-5p mimics.The difference was statistically significant(P<0.05).Conclusion:Shenling Baizhu Powder may inhibit the invasion of LoVo cells through Linc01615/miR-491-5p,promote apoptosis and block the cell cycle in G0/G1 phase.
