摘要
[目的]制备具有良好线性的蔗糖密度梯度溶液。[方法]利用溶液梯度混合仪和蠕动泵组装成新型的蔗糖密度梯度溶液制备器,用该制备器制备50%~0%和50%~10%等不同蔗糖密度梯度溶液,然后利用电脑核酸蛋白检测仪检测所制备的蔗糖密度梯度溶液的光密度,并对蔗糖密度溶液的线性进行评估;比较自制的蔗糖密度梯度溶液制备器与经典的Biocomp全自动密度梯度制备仪制备的蔗糖密度梯度溶液的优劣。[结果]自制的蔗糖密度梯度溶液制备器制备的50%~0%和50%~10%蔗糖密度梯度溶液的光密度与蔗糖密度的决定系数(R^(2))都在0.98以上;Biocomp全自动密度梯度制备仪制备的50%~0%蔗糖密度梯度溶液的光密度与蔗糖密度的决定系数(R^(2))为0.95~0.97。[结论]自制的蔗糖密度梯度溶液制备器制备的蔗糖密度梯度溶液优于Biocomp全自动密度梯度制备仪制备的蔗糖密度梯度溶液。
[Objective]To prepare sucrose density gradient solution with good linearity.[Method]A new sucrose density gradient solution preparator was assembled by utilizing a solution gradient mixer and a Peristaltic pump.This device was utilized to prepare 50%-0%and 50%-10%sucrose density gradient solutions,which were then analyzed by using computer nucleic acid protein detector.Additionally,the linearity of the sucrose density gradient solution prepared by the self-made preparator was compared to that of the classic Biocomp automatic density gradient separation system.[Result]The coefficient of determination(R-squared)of the 50%-0%and 50%-10%sucrose density gradient solutions prepared by the self-made sucrose density gradient solution preparator was found to be above 0.98.In comparison,the coefficient of determination(R-squared)of the 50%-0%sucrose density gradient solution prepared by the Biocomp automatic density gradient separation system was found to be between 0.95-0.97.[Conclusion]The linearity of the sucrose density gradient solution prepared by the self-made sucrose density gradient solution preparation device is superior to that prepared by the Biocomp automatic density gradient separation system.
