摘要
目的分析采用密度梯度离心进行精液处理时,不同离心力和离心时间对精子回收率的影响。方法选取2019年6月至2020年12月在邢台市第三医院行精液常规检测的84例不育患者作为研究对象。其中36例精子浓度低于15×10^(6)/mL而精子活动率正常(低浓度患者),48例精子活动率低于40%而精子浓度正常(低活动率患者)。按照密度梯度离心操作时的不同离心条件,将离心力300×g、离心时间20min处理的精液纳入A组,400×g、20min处理的纳入B组,500×g、10min处理的纳入C组,分析比较各组精子处理后的精子情况。结果低浓度患者处理后B组和C组的精子浓度及回收率均高于A组,差异具有统计学意义(P<0.05);而B组和C组的回收率比较,差异无统计学意义(P>0.05);但C组的精子活动率明显低于A组和B组,差异具有统计学意义(P<0.05)。低活动率患者处理后B组精子浓度、精子活动率及精子回收率均高于A组,差异具有统计学意义(P<0.05);B组活动率及精子回收率高于A组,差异具有统计学意义(P<0.05);但是C组精子活动率及回收率与B组、A组比较,差异无统计学意义(P<0.05)。结论不同离心条件会对精子的浓度、活动率及回收率有一定影响,应根据精液情况选择合适的离心条件,从而提高优化处理的效果。
Objective To analyze the influence of centrifugal force and time on sperm recovery during semen processing. Methods 84 infertility patients who underwent routine semen testing in the Third Hospital of Xingtai from June 2019 to December 2020 were selected as the research subjects. Among them, 36 cases had normal sperm motility with sperm concentration lower than 15×10^(6)/mL(low concentration patients), and 48 cases had normal sperm motility with sperm concentration lower than 40%(low motility patients). According to different centrifugal conditions of density gradient centrifugation, semen treated with centrifugal force of 300×g and centrifugation time of 20 min was included in group A, and semen treated with centrifugal force of 400×g and centrifugation time of 20 min was included in group B, the semen treated with 500×g and 10 min was included in group C, and the sperm condition of each group after sperm treatment was analyzed and compared. Results After treatment of low-concentration patients, the sperm concentration and recovery rates of group B and C were higher than those of group A(P<0.05);while the recovery rates of group B and C were not statistically significant(P>0.05);but the sperm motility rate of group C was significantly lower than that of group A and group B(P<0.05). After treatment of patients with low activity rate, the sperm concentration, sperm motility rate and sperm recovery rate of group B were higher than those of group A(P<0.05);the motility rate and sperm recovery rate of group B were higher than that of group A(P<0.05);however, the sperm motility rate and recovery rate of group C were not statistically significant(P<0.05) compared with group B and group A. Conclusions Different centrifugation conditions will have a certain impact on the concentration, activity rate and recovery rate of sperm. Appropriate centrifugation conditions should be selected according to the semen condition to improve the effect of optimized treatment.
