摘要
研究旨在丰富慢羽基因型的分子标记并探究慢羽基因型与产蛋量的相关关系。试验基于2个慢羽母鸡群体京红1号父母代种鸡母本和欣华鸡母本鉴定慢羽基因型的种类,采用PCR扩增方法检测个体中融合基因dPRLR-dSPEF2和内源性逆转录病毒ev21基因的插入情况。结果显示:在两个慢羽母鸡群体中存在6种不同的慢羽基因型,当个体存在ev21基因(OR)或融合基因dPRLR-dSPEF2中的任意一种或两种时均表现为慢羽,当无ev21基因(UR)且融合基因dPRLR-dSPEF2也不存在时,表现为快羽;11个其他品种快慢羽基因型频率统计发现不同的品种快慢羽基因型频率存在差异。个体中携带融合基因dPRLR-dSPEF2时,其产蛋量会显著高于不携带者(P<0.05);与仅携带ev21基因(OR突变)的个体相比,同时携带ev21基因(OR突变)和正常UR区域的产蛋量有显著升高(P<0.05)。研究表明,ev21插入和融合基因并非鸡快慢羽的唯一关联突变,且京红1号父母代群体中慢羽基因型间存在显著的产蛋量差异。
In this study,six different late-feathering genotypes in two late-feathering populations were identified to enrich the molecular markers of late-feathering and explore the correlation between late-feathering genotypes and egg production,in-cluding parental generation of Jinghong No.1 and Xinhua chickens.PCR amplification technology was used to detect insertion of ev21 and fusion gene dPRLR-dSPEF2 existed.The results were as follows:A total of six different late-feathering genotypes were found in two late-feathering populations.When either ev21 gene insertion(OR)or the fusion gene dPRLR-dSPEF2 existed,the individuals would exhibit late-feathering.Otherwise,neither ev21 gene insertion(UR)nor the fusion gene dPRLR-dSPEF2 exist-ed,the individuals would exhibit fast-feathering.The frequencies of late-feathering genotypes based on other 11 breedsfound dif-ference of those frequencies were in different breeds.And when the fusion gene dPRLR-dSPEF2 was present,the egg production was significantly higher than that of the non-existent.Comparing with the individuals only carrying OR mutation,the egg produc-tion of the individuals carrying UR mutation or both carrying OR and UR were significantly higher.In summary,the results indi-cated that ev21 gene insertion and fusion genes were not the only associated mutation of fast-feathering in chicken,and signifi-cant difference of egg production existed in different late-feathering genotypes.
