摘要
植物糖外排蛋白(SWEET)是新发现的一类糖转运体,影响植物的生长发育,但其在葡萄中的研究却鲜有报道。本研究对葡萄SWEETs基因家族(命名为VvSWEETs)进行全基因组鉴定和生物信息分析;分析VvSWEETs在野生型(有核)和突变型(无核)酿酒葡萄中的表达量,以及其在‘无核翠宝’葡萄胚挽救过程中的表达水平。结果显示,共鉴定到16个VvSWEETs,根据进化关系分为4个亚家族。VvSWEETs启动子区域包含大量激素响应类元件、植物发育类元件、胁迫响应类元件和光响应元件。通过对野生型和突变型酿酒葡萄VvSWEETs的特异表达研究,发现突变型酿酒葡萄胚珠中VvSWEET1、VvSWEET7和VvSWEET10表达下调。qRT-PCR分析显示,胚挽救过程中VvSWEET1、VvSWEET2a和VvSWEET10在‘无核翠宝’胚珠中表达显著上调,其中VvSWEET2a和VvSWEET10表达量与胚珠蔗糖和还原糖含量呈显著正相关,表明VvSWEETs可能通过上调无核葡萄胚珠中糖含量,促进其发育。本研究对葡萄SWEETs基因家族进行了全面的鉴定与分析,为了解其在葡萄中的功能和无核葡萄离体胚珠发育中的作用提供了理论依据。
Plant sugar efflux protein(SwEET)is a newly discovered sugar transporter,which affects plant growth and development,but its research in grape is rarely reported.In this study,the genome-wide identi-fication and bioinformatics analysis of grape SWEETs(named VvSWEETs)gene family were carried out.The expression of VvSWEETs in wild-type(seeded)and mutant(seedless)wine grape,as well as its ex-pression of in'Cuibao Seedless'grape during embryo rescue were analyzed.The results showed that 16 VvSWEETs were identified,which were divided into 4 subfamilies according to evolutionary relationships.The promoter region of VvsWEETs contains a large number of hormone responsive elements,plant developmental elements,stress responsive elements and light responsive elements.Through the study on the specific expression of VvSWEETs in wild-type and mutant wine grape,it was found that the expression of VvSWEET1,VvSWEET7 and VvSWEET10 were down-regulated in the ovule of mutant wine grape.qRT-PCR analysis showed that the expression of VvSWEET1,VvSWEET2a and VvSWEET10 were significantly up-regulated in the ovules of'Cuibao Seedless'during embryo rescue.The expression of VvSWEET2a and VvSWEET10 were positively correlated with the content of sucrose and reducing sugar in ovule,indicating that VvSWEETs may promote the development of seedless grape by up-regulating the sugar content in the ovule.In this study,the SwEETs gene family was comprehensively identified and analyzed in grape,which provided a theoretical basis for understanding its functions in grape and its roles in development of in vitro ovule of seedless grape.
