载淫羊藿苷和盐酸米诺环素明胶微球/骨水泥的构建及其性能研究

Construction and properties of calcium phosphate cement with icariin and minocycline hydrochloride-loaded gelatin microspheres

摘要 目的构建负载淫羊藿苷(ICA)和盐酸米诺环素(MH)的明胶微球/骨水泥(GMs/CPC)材料并对其性能进行研究,为种植体周围炎的治疗提供一种骨替代材料。方法制备GMs及负载ICA的GMs(ICA)、负载MH的GMs(MH)和同时负载两种药物的GMs(ICA+MH),将GMs(ICA+MH)与CPC复合构建CPC/GMs(ICA+MH)材料,将GMs(ICA)和GMs(MH)按照质量比1∶1的比例混合后与CPC复合构建CPC/GMs(ICA)+GMs(MH)材料。通过扫描电镜实验观察材料表面形态,X射线衍射实验分析材料物相。将各组材料浸提液作用于骨髓间充质干细胞(BMSCs),通过CCK-8实验检测材料浸提液对BMSCs增殖作用的影响,通过茜素红染色及钙含量定量分析实验检测材料浸提液及含浸提液的矿化液对BMSCs矿化作用的影响。结果扫描电镜观察显示,CPC/GMs、CPC/GMs(ICA+MH)和CPC/GMs(ICA)+GMs(MH)中的明胶微球被CPC基质包裹并随机散在分布于骨水泥中;X射线衍射分析结果显示以上3种材料中明胶微球与CPC结合后并未发生化学反应产生新的物质。CCK-8实验结果显示,BMSCs在材料浸提液中培养7 d后,CPC/GMs(ICA)+GMs(MH)组BMSCs的增殖率显著升高,与CPC/GMs组和CPC/GMs(ICA+MH)组比较差异具有统计学意义(P<0.05)。BMSCs在材料浸提液及含浸提液的矿化液中培养21 d后,CPC/GMs(ICA)+GMs(MH)组矿化能力显著高于CPC/GMs组和CPC/GMs(ICA+MH)组(P<0.05)。结论CPC/GMs(ICA+MH)和CPC/GMs(ICA)+GMs(MH)材料对BMSCs的成骨分化均具有促进作用,其中CPC/GMs(ICA)+GMs(MH)的作用较强是一种优选的骨替代材料。 Objective To construct calcium phosphate cements(CPCs)with icariin(ICA)and minocycline hydrochloride(MH)loaded gelatin microspheres(GMs)and study their properties for providing a bone substitute material in the treatment of peri-implantitis.Methods GMs,GMs loaded with ICA,GMs loaded with MH and GMs loaded with both drugs were prepared.The CPC/GMs(ICA+MH)was constructed by combining GMs(ICA+MH)with CPC,while the CPC/GMs(ICA)+GMs(MH)was constructed by combining the mixture of GMs(ICA)and GMs(MH)in a mass ratio of 1∶1 with CPC.The surface morphology of the materials was observed by scanning electron microscopy(SEM),and the phase was analyzed by X-ray diffraction.The effects of the material extracts on the proliferation of BMSCs were detected by CCK-8 assay,the effects of the material extracts and the mineralizing solution containing the extracts on the mineralization of BMSCs were detected by Alizarin red staining and quantitative analysis of calcium content.Results SEM showed that GMs were randomly dispersed and enfolded with CPC in CPC/GMs,CPC/GMs(ICA+MH),and CPC/GMs(ICA)+GMs(MH),and X-ray diffraction analysis showed that no new substance was formed after the combination of all kinds of GMs and CPC.The results of CCK-8 assay showed that the proliferation of BMSCs was significantly increased in CPC/GMs(ICA)+GMs(MH)group after cultured with the material extracts for 7 d,and it was higher than that in CPC/GMs group and CPC/GMs(ICA+MH)group(P<0.05).After BMSCs were cultured in material extracts and mineralizing solutions containing the extracts for 21 d,the mineralization of BMSCs was statistically higher in CPC/GMs(ICA)+GMs(MH)group than that in CPC/GMs group and CPC/GMs(ICA+MH)group(P<0.05).Conclusion CPC/GMs(ICA+MH)and CPC/GMs(ICA)+GMs(MH)can promote the osteogenic differentiation of BMSCs.CPC/GMs(ICA)+GMs(MH)has a stronger effect,and can be a preferred bone substitute material.
