摘要
目的 测定桑叶药材芦丁含量及高效液相(HPLC)特征图谱,结合多种化学计量分析方法综合评价桑叶药材质量。方法 采集34批不同产地的桑叶药材HPLC特征图谱,通过相似度评价,化学模式识别和AHPCRITIC混合加权法对桑叶特征成分进行差异性研究。另测定桑叶芦丁含量,评价其与桑叶药材色度值的相关性。结果 34批桑叶样品确定了7个共有峰,指认了其中3个成分;各批样品HPLC特征图谱与对照图谱的相似度均>0.903,聚类分析和主成分分析结果表明34批桑叶样品可大致分成3类,正交偏最小二乘判别分析表明3(隐绿原酸),5,1(新绿原酸),7号色谱峰为区分桑叶质量差异的标志物,AHP-CRITIC混合加权法结果显示综合评分>42分的桑叶药材质量较优。桑叶芦丁含量与色度值的相关性分析结果表明偏亮黄色的桑叶芦丁含量较高。结论 本研究方法能较全面、真实地反映桑叶药材的内在质量,可为桑叶药材的质量控制提供参考。
Objective To determine the content of rutin in Mori Folium and its characteristic HPLC chromatogram,and evaluate its quality comprehensively by combination of various stoichiometric analysis methods.Methods The characteristic HPLC chromatograms of 34 batches of Mori Folium herbs from different producing areas were collected,and differential study of characteristic components of Mori Folium were conducted through similarity evaluation,chemical pattern recognition,and AHP-CRITIC mixed weighting method.In addition,the content of rutin was determined and its correlation with the chromatic value of Mori Folium herbs was evaluated.Results 7 common peaks were confirmed in 34 batches of samples,among which 3 peaks were identified.Similarity analysis showed that the similarity between the characteristic HPLC chromatograms of each batch of sample and the reference was higher than 0.903.The results of cluster analysis and principal component analysis showed that 34 batches of Mori Folium samples could be roughly divided into 3 categories.Orthogonal partial least squares discriminant analysis showed that peaks 3(cryptochlorogenic acid),5,1(neochlorogenic acid)and 7 were markers to distinguish the quality difference of Mori Folium samples.The results of AHP-CRITIC mixed weighting method showed that the quality of Mori Folium herbs with comprehensive score>42 were better.The correlation analysis between rutin content and chromatic value of Mori Folium herbs showed that the content of rutin in bright yellow samples was higher.Conclusion This method can comprehensively and truly reflect the inner quality of Mori Folium herbs,and can provide reference for the quality control of Mori Folium herbs.
