HPLC法测定葫芦七不同部位中7种倍半萜的含量

摘要 [目的]建立HPLC法测定葫芦七不同部位(根、茎、叶)中7种倍半萜类化合物的含量。[方法]采用Kromasil C_(18)(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-水(40∶60);柱温35℃;检测波长240 nm;进样体积10μL。[结果]7种被测成分在测定质量浓度范围内线性关系良好(R^(2)≥0.9990);平均回收率在95.18%~97.61%,RSD在0.41%~1.60%;被检测的7种倍半萜类成分在葫芦七根、茎、叶不同部位中的含量差异明显,根中所含化合物的种类相对较多,含量相对较高。[结论]该方法快速、简便、准确,可用于葫芦七药材的质量控制。 [Objective]To establish a HPLC method determinate seven sesquiterpenes in different parts of Ligularia fischeri.[Method]Chromatographic column was Kromasil C_(18)(4.6 mm)×250 mm,5μm),mobile phase was acetonitrile water(40∶60);column temperature was 35℃;detection wavelength was 240 nm;injection volume was 10μL.[Result]The linearity of seven sesquiterpenes was good in the range of their mass concentration(R^(2)≥0.9990);the average recovery rates were 95.18%-97.61%,RSDs were 0.41%-1.60%.The content of seven sesquiterpenoids detected in different parts of the root,stem and leaf of Ligularia fischeri were significantly different.The content and kinds of compounds in roots were higher than that in other parts.[Conclusion]The method was fast,simple and accurate,and can be used for the quality control of Ligularia fischeri.
