摘要
目的探究周期素依赖性蛋白激酶10(CDK10)在肝细胞癌(HCC)组织内的表达及与患者临床特征和预后的关系。方法采用免疫组化技术检测在HCC组织芯片中CDK10的表达情况;采用卡方分析比较CDK10表达差异与肝细胞癌患者各临床参数之间的关联,并结合Kaplan-Meier生存分析和COX比例风险模型评价CDK10的表达与患者预后的相关性。结果CDK10蛋白在HCC组织中阳性表达于细胞质,癌组织中的表达(32.98%)显著低于癌旁组织(59.30%)(P<0.05)。CDK10高表达与组织学分级、肿瘤分期、TNM分期、肿瘤大小、血清AFP水平均呈正相关(P<0.05)。CDK10高表达组肝细胞癌患者的术后生存时间明显高于CDK10低表达组肝细胞癌患者(P<0.05);COX多因素分析提示,CDK10表达水平与组织学分级可作为影响肝细胞癌患者生存的独立预后指标(P<0.05)。结论CDK10可作为一种抑癌基因低表达于HCC患者的肿瘤组织,有望成为肝癌早期诊断的分子标志物和判断预后的参考指标。
Objective To explore the expression of cyclin-dependent kinase 10(CDK10)in hepatocellular carcinoma(HCC)tissue and its relationship with clinical characteristics and prognosis of patients.Methods The expression of CDK10 in HCC tissue microarray was detected by immunohistochemical technique.The correlation between CDK10 expression and clinical parameters of HCC patients was analyzed byχ2 analysis.The correlation between CDK10 expression and prognosis of HCC patients was evaluated by Kaplan-Meier survival analyses and COX proportional hazards model.Results CDK10 was positively expressed in the cytoplasm of HCC tissues and its positive expression rate of CDK10 protein in hepatocellular carcinoma tissues and para-carcinoma tissues were 32.98%and 59.30%(P<0.05).The expression level of CDK10 was significantly correlated with histological grade,tumor stage,TNM stage,tumor size and serum AFP level.The postoperative survival time of patients with hepatocellular carcinoma in CDK10 high expression group was significantly higher than that in CDK10 low expression group(P<0.05).COX multivariate analysis showed that CDK10 expression level and histological grade could be used as independent prognostic indicators affecting the survival of patients with hepatocellular carcinoma(P<0.05).Conclusion CDK10 can be regarded as an anti-oncogene with low expression in neoplasm tissues of hepatic carcinoma patients,which is expected to be used as a molecular biomarker for early diagnosis of HCC and an indicator for prognosis.
