润楠叶片多糖提取工艺、结构表征与生物活性研究 被引量:1

摘要 为探究润楠叶片多糖的最佳提取工艺和结构表征,分析其体外抗氧化及抗肿瘤细胞活性。该文以产自湖北省恩施市的润楠叶片为材料,通过单因素试验探究体系pH、超声温度、超声功率及超声时间对润楠叶片多糖得率的影响,正交试验设计优化超声辅助复合酶(纤维素酶、果胶酶)提取润楠叶片多糖工艺;采用傅里叶红外光谱法和扫描电镜分析润楠叶片多糖结构;进一步采用1-苯基-3-甲基-5-吡唑啉酮柱前衍生高效液相色谱(PMP-HPLC)法测定其单糖组成;最后,采用体外抗氧化实验和细胞实验探究其生物活性。实验结果表明润楠叶片多糖的最佳提取工艺为体系pH为5、超声温度50℃、超声功率250 W、超声时间20 min,多糖得率达到7.00%以上;润楠叶片多糖红外光谱图呈现出多糖的典型特征吸收峰;扫描电镜图呈结构疏松不规则的块状形态;单糖组成分析表明润楠叶片多糖主要由甘露糖、阿拉伯糖、半乳糖醛酸、葡萄糖和阿拉伯糖等构成的水溶性杂多糖;体外抗氧化、细胞抗肿瘤试验表明润楠叶片多糖具有良好的生物活性,在保健、生物食品与药品等方面具有开发潜能。 In order to investigate the optimal extraction process and structural characterization of Machilus nanmu leaf polysaccharides,and their antioxidant and anti-tumor cell activities in vitro.In this study,using M.nanmu leaf produced in Enshi City,Hubei Province as the material,the effects of system pH,ultrasonic temperature,ultrasonic power and ultrasound time on the polysaccharide yield of M.nanmu leaf were explored by one-factor experiments,and orthogonal experimental design optimized the polysaccharide extraction process of ultrasonic auxiliary compound enzymes(cellulase and pectinase)from M.nanmu leaf.Further analysis of polysaccharide structure of M.nanmu leaf by Fourier infrared spectroscopy and scanning electron microscopy;The monosaccharide composition of 1-phenyl-3-methyl-5-pyrazolinone pre-column derived high performance liquid chromatography(PMP-HPLC)was used.Experimental results showed that the optimum extraction process of M.nanmu leaf polysaccharide were pH=5,ultrasonic temperature 50℃,ultrasonic power 250 W,ultrasonic time 20 min,and the yield of polysaccharide was more than 7.00%.The infrared spectrum showed the typical characteristic absorption peak of polysaccharide.Scanning electron microscope showed loose and irregular block shape.The analysis of monosaccharide composition showed that M.nanmu leaf polysaccharide was a water-soluble heteropolysaccharide mainly composed of mannose,arabinose,galacturonic acid,glucose and arabincose.In vitro antioxidant and cell antitumor tests showed that M.nanmu leaf polysaccharide had biological activity and had development potential in health care,biologicalfood and medicine.
