箬竹叶酸性多糖对糖尿病视网膜病变小鼠的影响及机制研究

摘要 为探讨箬竹叶酸性多糖对糖尿病视网膜病变小鼠的影响及机制,通过链脲佐菌和高糖高脂饮食建立小鼠糖尿病模型,用不同剂量箬竹叶酸性多糖(500、300、100 mg/kg)对其进行连续4周的干预治疗。通过检测小鼠空腹血糖、HbA1c和AGEs含量以研究其降血糖能力,进行视网膜电图和光学相干断层成像对视网膜生理指标检测,并测定视网膜组织的抗氧化能力和炎症相关指标含量,测定其抗氧化相关基因和细胞凋亡相关基因表达量。结果表明箬竹叶酸性多糖可以降低小鼠空腹血糖、HbA1c和AGEs含量,具有一定的降血糖能力。在抗氧化上,箬竹叶酸性多糖可以通过增加SOD、CAT和GSH-Px酶活性提高T-AOC活性以降低MDA含量。在炎症上,箬竹叶酸性多糖可以抑制炎症因子IL-1β、TNF-α和IL-6的释放,还可降低VEGF含量以免视网膜组织血管增生。箬竹叶酸性多糖通过Nrf2-ARE通路上调抗氧化酶类表达量,以减轻氧化应激反应,下调NF-κB途径涉及的炎症反应、细胞凋亡和血管增生,进而缓解高血糖视网膜病变。箬竹叶酸性多糖是一种利于高血糖患者的潜在功能性食品添加剂。 In order to explore the effect of acidic polysaccharide from Indocalamus leaves on diabetes retinopathy in mice and its mechanism,a diabetes mice model was established by streptozotocin and high sugar and high fat diet,and different doses of acidic polysaccharide(500,300,100 mg/kg)from Indocalamus leaves were used to treat it for four consecutive weeks.The hypoglycemic ability of mice was studied by detecting the contents of fasting blood glucose,HbA1c and AGEs.The physiological indexes of retina were detected by electroretinogram and optical coherence tomography,and the antioxidant capacity and inflammatory related indexes of retinal tissue were measured.The expression of antioxidant related genes and apoptosis related genes were measured.The results showed that acidic polysaccharide from Indocalamus leaves could reduce the fasting blood glucose,HbA1c and AGEs content in mice,and had certain hypoglycemic ability.In terms of antioxidation,acid polysaccharide from Indocalamus leaves could increase T-AOC activity and reduce MDA content by increasing SOD,CAT and GSH-Px enzyme activities.In inflammation,acid polysaccharide from Indocalamus leaves could inhibit the release of inflammatory factor of IL-1β,TNF-αand IL-6,and reduce the content of VEGF to avoid retinal vascular hyperplasia.Acid polysaccharide from Indocalamus leaves could alleviate hyperglycemic retinopathy to up regulate the expression of antioxidant enzymes through Nrf2-ARE pathway to reduce oxidative stress reaction and down regulate inflammatory reaction,cell apoptosis and vascular proliferation involved in the NF-κB pathway.Acid polysaccharide from Indocalamus leaves could be a potential functional food additive for diabetic.
