摘要
以柑橘杂交种子为试材,研究培养介质和培养基添加植物生长调节剂和无机盐对柑橘种子发芽和胚根生长的影响,统计不同处理对柑橘杂交苗生长状态、嫁接成活率和嫁接效率的影响,以期筛选出适合柑橘苗生长的培养介质。结果表明,琼脂培养的柑橘种子出芽率、胚根长、胚根生长量分别为75.56%、13.64 mm、12.81 mm,比蛭石和滤纸分别提高了25.93%、23.22%、122.78%和209.17%、81.62%、64.44%。从形态观察可知,琼脂培养柑橘杂交种子能随时观察种胚生长状态,种胚直、生长状况良好,在非无菌环境下操作,未出现污染现象,反复回栽不伤胚根,不影响胚根的继续生长;蛭石培养种子胚根生长不整齐,回栽严重影响胚根的继续生长;滤纸培养胚根弯曲严重,不利于后续嫁接操作。琼脂+植物生长调节剂+无机盐培养的柑橘种子出芽率、胚根长、胚根粗、胚根生长量分别为91.11%、16.48 mm、2.08 mm、16.71 mm,比琼脂处理分别提高了20.58%、20.82%、40.54%、30.44%,比琼脂+植物生长调节剂处理分别提高了24.25%、42.19%、23.08%、31.06%,比琼脂+无机盐处理分别提高了25.05%、36.65%、14.92%、36.52%,说明琼脂添加植物生长调节剂和无机盐培养的柑橘种子发芽整齐、生长速度快、胚根粗壮。综合来看,琼脂培养基较适合柑橘种子嫁接苗的培养,添加植物生长调节剂和无机盐能提高柑橘种子发芽整齐度和促进胚根的增粗生长,有利于后续柑橘杂交苗的嫁接操作,可提高嫁接成活率和嫁接效率。
In this study,citrus hybrid seeds were used as the material.In order to screen out the culture medium suitable for citrus seedling growth,the effects of the addition of plant growth regulator and inorganic salt on citrus seeds germination and radicle growth were studied,and the effects of different treatments on the growth status,grafting survival rate and grafting efficiency of citrus hybrid seedlings were statistically calculated.The results showed that the germination rate,radicle length and radicle growth of citrus seeds cultured in agar were 75.56%,13.64 mm and 12.81 mm,which were 25.93%,23.22%,122.78%and 209.17%,81.62%,64.44%higher than vermiculite culture and filter paper culture,respectively.From the morphological observation,agar cultured citrus hybrid seeds can observe the growth state of the embryo at any time,the embryo is straight,the growth condition is good,it is operated in a non-sterile environment,there is no pollution phenomenon,and repeated replanting does not damage the radicle,which does not affect the continued growth of the radicle.The radicle growth of vermiculite culture seeds is untidy,and replanting seriously affects the continued growth of radicle.The radicle bending of filter paper culture is serious,which is unconducive to subsequent grafting operations.The germination rate,radicle length,radicle diameter,and radicle growth of citrus seeds cultured on agar+plant growth regulator+inorganic salt added were 91.11%,16.48 mm,2.08 mm,and 16.71 mm,respectively,which were 20.58%,20.82%,40.54%,and 30.44%higher than those treated with agar,24.25%,42.19%,23.08%,and 31.06%higher than those treated with agar+plant growth regulator,and 25.05%,36.65%,14.92%,and 36.52%higher than those treated with agar+inorganic salt,respectively.Explanation:citrus seeds cultured with agar supplemented with plant growth regulator and inorganic salt exhibit neat germination,fast growth rate,and robust radicles.The results show that agar medium is more suitable for the cultivation of citrus seed grafted seedlings,an
