摘要
本研究以黄瓜低霜霉威残留品系‘D0351’为材料,克隆了在霜霉威胁迫后上调表达的基因Cs PYCR,并采用生物信息学方法对该基因进行分析,以明确该基因在降低黄瓜果实内霜霉威残留过程中的作用机制,为下一步通过遗传转化试验鉴定基因功能提供理论基础。结果表明:该基因全长831 bp,编码276个氨基酸,属于PLN02688蛋白家族。该基因编码的蛋白质有可能为细胞质中的疏水蛋白质,无明显信号肽,无跨膜结构。与甜瓜(XM_008457466.2)同源性较高。该基因启动子中包括与乙烯、脱落酸等激素相关的顺式作用元件以及防御和应激反应相关的顺式作用元件,且含有多个与逆境胁迫相关的MYB家族蛋白转录因子结合位点,推测其可能在降低黄瓜霜霉威农药残留过程中发挥重要作用。
In this study,the up-regulated gene CsPYCR was cloned from the low propamocarb residue strains' D0351',which was stressed by propamocarb,and the gene was analyzed by bioinformatics methods to clarify the mechanism of the gene in the process of reducing the residue of propamocarb in cucumber fruit.It provides a theoretical basis for identifying gene function by genetic transformation test.The results showed that the gene was 831 bp long and encoded 276 amino acids and a PLN02688 domain.The protein encoded by this gene may be hydrophobic protein in cytoplasm,without obvious signal peptide and transmembrane structure.Evolutionary tree analysis showed that CsPYCR had the highest homology with Cucumis melo(XM_008457466.2).The core elements of promoter include cis-acting elements related to hormones such as ETH and ABA,cis-acting elements related to defense and stress response.It also contains a number of MYB family protein transcription factor binding sites related to stress,suggesting that they may play an important role in the process of reducing the residue of propamocarb pesticide in cucumber.
