星点设计响应面法优化参芪二皮冬茶提取工艺

摘要 目的:优化参芪二皮冬茶提取工艺。方法:结合处方中各药的质量标志物,采用紫外分光光度法测定总多糖含量,以HPLC法建立参芪二皮冬茶指纹图谱并进行共有峰的归属,选取阿魏酸、毛蕊异黄酮葡萄糖苷、橙皮苷、迷迭香酸、总多糖含量以及干膏率作为评价指标,通过层次分析法-熵权法计算权重系数;采用星点设计-响应面实验对提取时间、加水倍数进行考察,通过综合评分对提取工艺进行优化。在此基础上,采用单因素实验考察提取次数,优化工艺参数并进行验证实验。结果:优化结果为12倍水量煎煮2次,每次95 min。结论:该方法因其具备良好的可行性、稳定性,可为参芪二皮冬茶工业生产提供依据。 Objective:To optimize the extraction process of Shenqi Erpi Dong tea.Methods:Combined with the quality markers of each drug in the prescription,the total polysaccharide content was determined by ultraviolet spectrophotometry,and the fingerprint of Shenqi Erpi dong tea was established by HPLC method and the common peak was classified.Ferulic acid,calycosin-7-glucoside,hesperidin,rosmarinic acid,total polysaccharide content,and dry paste rate were selected as the evaluation indicators,and the weight coefficient was calculated by the AHP-entropy weight method.Star design-response surface test was used to investigate the extraction time and water addition multiple,and the extraction process was optimized by comprehensive scoring.Results:The optimal result was to decoction 2 times with 12 times water volume for 95 min each time.Conclusion:The method can provide a basis for the industrial production of Shenqi Erpi Dong tea due to its good feasibility and stability.
