摘要
介绍一种改良的适用于心血管效应机理研究中大鼠心肌组织灌注取材的新型实验装置及方法。其创新之处在于将离体心脏主动脉插管逆行灌注的原理与传统灌注方法相结合,设计出的这款新型装置可避免常规在体心脏灌流固定取材时应用主动脉夹及针头导致大鼠心肌组织夹伤破损的问题,且结构简单、易于操作。制作出的心肌组织切片完整、美观,镜下心肌组织结构清晰,荧光免疫组化染色效果理想。通过该装置及方法的应用,有利于针灸、药物或针药结合心血管效应机理研究相关的形态学观察及结果展示。
This article introduces an improved new experimental device and method suitable for perfusion sampling of rat myocardial tissue in the study of cardiovascular effect mechanisms.The innovation of this study is that it combines the principle of retrograde perfusion of the isolated heart aortic cannula with the traditional perfusion method,and the design of this new device can avoid the injury of rat heart from aortic clamp and needle during cardiac perfusion.Moreover,the device is simple and easy to operate.The myocardial tissue sections prepared by the new device are intact and beautiful,the myocardial tissue structure under the microscope is clear,and the fluorescent immunohistochemical staining effect is ideal.Through the application of this device and method,it is conducive for morphological observation and result display related to acupuncture,medicine or acupuncture combined with cardiovascular effect mechanism research.
