小儿清咽颗粒中8个成分的含量测定及主成分分析 被引量:1

摘要 目的建立同时测定小儿清咽颗粒中哈巴苷、(R,S)-告依春、连翘酯苷A、菊苣酸、连翘苷、牛蒡苷、哈巴俄苷和丹皮酚含量的高效液相色谱方法。方法采用Merck Purospher C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈(A)-0.05%磷酸水溶液(B),梯度洗脱,流速0.8 mL·min^(-1),柱温30℃,检测波长210 nm(哈巴苷、连翘酯苷A、连翘苷、牛蒡苷、哈巴俄苷、丹皮酚)、245 nm[(R,S)-告依春]、327 nm(菊苣酸)。再用SPSS 20.0统计软件对结果进行聚类分析和主成分分析。结果各待测组分分离度良好;哈巴苷、(R,S)-告依春、连翘酯苷A、菊苣酸、连翘苷、牛蒡苷、哈巴俄苷和丹皮酚8个成分在相应的范围内均与峰面积呈良好的线性关系,相关系数(r)均不低于0.9997,低、中、高浓度的平均加样回收率均在96.9%~100.1%,RSD均小于1.3%。结论该方法完善了现行标准,可用于小儿清咽颗粒的质量控制。 Objective To establish an HPLC method to simultaneously determinate harpagide,(R,S)-goitrin,forsythoside A,chicoric acid,forsythin,arctiin,harpagoside and paeonol in Xiaoer Qingyan granules.Methods The samples were separated on a Merck Purospher C18 column(250 mm×4.6 mm,5μm)by a gradient elution with acetonitrile(A)and 0.05%phosphoric acid(B)as the mobile phase at the flow rate of 0.8 mL·min^(-1).The column temperature was 30℃.The eluate was detected by multiple wavelength at 210 nm for harpagide,forsythoside A,forsythin,arctiin,harpagoside,and paeonol;245 nm for(R,S)-goitrin,and 327 nm for chicoric acid.The principal component analysis and cluster analysis were conducted for the content determination by SPSS 20.0.Results Excellent chromatographic separation was achieved with good linearity(r≥0.9997)within the linear range of harpagide,(R,S)-goitrin,forsythoside A,chicoric acid,forsythin,arctiin,harpagoside and paeonol,respectively.The average recovery of low,medium and high concentrations was within 96.9%~100.1%,with RSD less than 1.3%.Conclusion The method improves the existing standards,which is suitable for the quality control of Xiaoer Qingyan granules.
