补肾调泡法联合维生素D对多囊卵巢模型大鼠卵巢抗苗勒氏管激素Ⅱ型受体表达的影响研究

摘要 目的 探讨补肾调泡法联合维生素D对多囊卵巢(PCOS)模型大鼠卵巢抗苗勒氏管激素Ⅱ型受体表达的影响。方法 30只SD雌性大鼠随机选取6只作为空白组,剩余24只采用来曲唑-羧甲基纤维素钠溶液灌胃法构建PCOS大鼠模型,并在成功后设立模型组、中药组(补肾调泡法)、维生素D组、联合组(补肾调泡法+维生素D),每组6只。实验33d后处死大鼠收集血标本及卵巢组织,测定血清激素水平、AGEs、25(OH)D、钙、磷、AMH,并对卵巢组织行HE染色,检测卵巢AMH及AMHRⅡ表达。结果 联合组、维生素D组、中药组的LH、FSH、T、AMH水平均低于模型组,且联合组各项指标低于维生素D组、中药组,维生素D组、中药组各项指标高于空白组,维生素D组E2明显升高,中药组的AGEs水平较高(P<0.05),但联合组与空白组相当(P>0.05);联合组、维生素D组、中药组的卵巢AMH、AMHRⅡ低于模型组(P<0.05),但联合组与空白组相当(P>0.05);联合组、维生素D组、中药组的25(OH)D、钙、磷水平高于模型组(P<0.05),但联合组与空白组相当(P>0.05)。结论 补肾调泡法联合维生素D对PCOS模型大鼠卵巢AMHRⅡ表达有抑制作用,通过抑制AMH信号通路,提高25(OH)D水平,调节性激素水平,降低AGEs,可促进卵泡发育和成熟,改善卵巢功能。 Objective To investigate effect of kidney tonifying and bubble regulating therapy combined with vitamin D on expression of anti mullerian hormone typeⅡreceptor in ovaries of polycystic ovary(PCOS)model rats.Methods The paper chose 6 SD female rats of 30 as blank group and PCOS rat model was constructed with remaining 24 rats successfully based on gavage of letrozole carboxymethyl cellulose sodium solution,and divided them into model group,traditional Chinese medicine group(kidney tonifying and bubble regulating method),vitamin D group and combination group(kidney tonifying and bubble regulating method+vitamin D).After 33 days of experiment,rats were euthanized to collect blood samples and ovarian tissue.Serum hormone levels,AGEs,25(OH)D,calcium,phosphorus and AMH were measured and ovarian tissue was stained with HE to detect expression of AMH and AMHRⅡin ovaries.Results Levels of LH,FSH,T and AMH in combination group,vitamin D group and traditional Chinese medicine group were lower than model group,those of combination group were lower than vitamin D group and traditional Chinese medicine group.Those of vitamin D group and traditional Chinese medicine group were higher than blank group,E2 in vitamin D group was significantly increased.AGEs Level in traditional Chinese medicine group was higher(P<0.05),but that of combination group was comparable to blank group(P>0.05).Ovarian AMH and AMHRⅡlevels in combination group,vitamin D group and traditional Chinese medicine group were lower than model group(P<0.05),but those of combination group were comparable to the blank group(P>0.05).Levels of 25(OH)D,calcium and phosphorus in combination group,vitamin D group and traditional Chinese medicine group were higher than model group(P<0.05),but those of combination group were comparable to the blank group(P>0.05).Conclusion Combination of tonifying kidney and regulating follicles method and vitamin D has inhibitory effect on expression of AMHRⅡin ovaries of PCOS model rats,can inhibite AMH signaling pathway,increase
