摘要
目的 观察续筋接骨液对兔肩袖损伤修复术后腱骨界面组织中磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)信号通路的影响,探讨续筋接骨液促进腱骨愈合的可能机制。方法 从15只新西兰大白兔中随机选取5只作为空白组,剩余10只制备兔肩袖损伤修复模型,将造模后兔随机分为模型组和续筋接骨组各5只。空白组、模型组兔给予生理盐水灌胃,续筋接骨组兔给予56 mg/kg续筋接骨液灌胃,均2次/d,连续灌胃8周。灌胃结束后,HE染色观察腱骨界面组织形态,Western blot法检测腱骨界面组织中p-PI3K、p-Akt蛋白表达情况,RT-qPCR法检测腱骨界面组织中PI3K、Akt mRNA表达情况。结果 与空白组比较,模型组兔腱骨连接不完全,胶原维排列紊乱,腱骨组织中p-PI3K、p-Akt蛋白及PI3K、Akt mRNA相对表达量均明显增高(P均<0.05)。与模型组比较,续筋接骨组兔腱骨连接紧密,胶原纤维排列有序,腱骨组织中p-PI3K、p-Akt蛋白及PI3K、Akt mRNA相对表达量均明显均降低(P均<0.05)。结论 续筋接骨液可以促进兔肩袖损伤修复术后腱骨愈合,作用机制可能与抑制PI3K/Akt信号通路活化有关。
Objective It is to observe the effect of Xujin Jiegu liquid on the phosphoinositide 3-kinase(PI3K)/protein kinase B(Akt)signaling pathway of tendon-bone interface in rabbit rotator cuff injury repair models,and to explore the possible mechanism of Xujin Jiegu liquid in promoting tendon-bone healing.Methods Fifteen New Zealand white rabbits were selected,in which 5 rabbits were randomly selected as the blank group,and the remaining rabbits were used to prepare rabbit rotator cuff injury repair models.The modeled rabbits were randomly divide model group and Xujin Jiegu liquid group,with 5 rabbits in each group.The rabbits of the blank group and model group were gavaged with physiological saline,while the ones of the Xujin Jiegu liquid group were gavaged with Xujin Jiegu liquid,all twice daily,continuously treated for 8 weeks.At the end of gavage,the morphology of the tendon-bone interface was observed by HE staining,the expressions of p-PI3K and p-Akt proteins in the tendon-bone interface were detected by Western Blot,and the expression of PI3K and Akt mRNA in the tendon-bone interface were detected by RT-qPCR.Results Compared with the blank group,incomplete tendon bone connection,disordered arrangement of collagenous fibers were found in the model group,and the relative expressions of p-PI3K,p-Akt proteins and PI3K,Akt mRNA in tendon-bone tissue were significantly increased(all P<0.05).Compared with the model group,the tendon bone connections were tight,the arrangement of collagenous fibers were order,and the relative expressions of p-PI3K,p-Akt proteins and PI3K,Akt mRNA in tendon-bone tissue were significantly decreased in the Xujin Jiegu liquid group(all P<0.05).Conclusion Xujin Jiegu liquid can promote tendon-bone healing in rabbit rotator cuff injury repair models,and its mechanism of action may be related to inhibiting the PI3K/Akt signaling pathway.
