基于TGR5/cAMP/PKA信号通路探讨疏肝、健脾疗法对肝损伤大鼠的作用及机制

Effects and mechanisms of therapy for soothing liver and invigorating spleen on liver-injury in rats based on TGR5/cAMP/PKA signaling pathway

摘要 目的 探讨疏肝疗法与健脾疗法对肝郁叠加肝损伤大鼠的作用及机制。方法 将30只雄性SD大鼠随机分为正常组、模型组、双环醇组、四逆散组、六君子汤组,每组6只。除正常组外,其余组大鼠均采用束缚叠加四氯化碳橄榄油溶液皮下注射的方法制备肝郁叠加肝损伤模型,造模同时模型组给予去离子水灌胃,双环醇组给予0.2 g/kg双环醇灌胃,四逆散组给予4.32 g/kg四逆散灌胃,六君子汤组给予六君子汤9.26 g/kg灌胃,连续灌胃42 d。末次灌胃结束后,全自动生化分析仪检测血清肝功能生化指标[碱性磷酸酶(ALP)、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总胆红素(TBil)、直接胆红素(DBil)]水平,RT-PCR法检测肝脏组织中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、环磷酸腺苷(cAMP)、蛋白激酶A(PKA)mRNA表达情况,免疫组化法检测肝脏、结肠组织中G蛋白偶联受体5(TGR5)阳性表达情况,AB-PAS染色观察结肠组织中杯状细胞数量。结果 与正常组比较,模型组血清ALP、ALT、AST、TBil、DBil水平和肝脏组织中IL-6 mRNA相对表达量、结肠组织中TGR5阳性表达平均光密度值均明显升高(P均<0.05),肝脏组织中cAMP、PKA mRNA相对表达量和肝脏组织中TGR5阳性表达平均光密度值均明显降低(P均<0.05),结肠组织中杯状细胞数量明显减少(P<0.05);与模型组比较,双环醇组、四逆散组血清ALP、ALT、AST、TBil、DBil水平及六君子汤组血清ALP、ALT、TBil、DBil水平,双环醇组、四逆散组肝脏组织中TNF-α、IL-6 mRNA相对表达量和各药物组结肠组织中TGR5阳性表达平均光密度值均明显降低(P均<0.05),四逆散组肝脏组织中cAMP、PKA mRNA相对表达量和双环醇组PKA mRNA相对表达量、六君子汤组cAMP mRNA相对表达量、各药物组肝脏组织中TGR5阳性表达平均光密度值均明显升高(P均<0.05),各药物组杯状细胞数量均明显增多(P均<0.05);与六君子汤组比� Objective It is to explore the effects and mechanisms of therapy for soothing liver and invigorating spleen on rats with liver depression combined with liver injury.Methods Thirty male SD rats were randomly divided into normal group,model group,bicyclol group,Sini Powder group,and Liujunzi Decoction group,with 6 rats in each group.Except for the normal group,the rats in the other groups were restrained and subcutaneously injected with carbon tetrachloride olive oil solution to prepare the models of liver depression with liver injury.At the same time as the model was being built,the model group was given deionized water by gavage,the bicyclol group was given 0.2 g/kg bicyclol by gavage,the Sini Powder group was given 4.32 g/kg Sini Powder by gavage,and the Liujunzi Decoction group was given 9.26 g/kg Liujunzi Decoction by gavage,all the groups were continuously gavaged for 42 days.After the last intragastric administration,the levels of serum liver function biochemical indicators[alkaline phosphatase(ALP),alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil),direct bilirubin(DBil)]were detected by a fully automatic biochemical analyzer,the expressions of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),cyclic adenosine monophosphate(cAMP)and protein kinase A(PKA)mRNA in liver tissue were detected by RT-PCR.the positive expression of G protein coupled receptor 5(TGR5)in liver and colon tissues were detected by immunohistochemistry,and the number of goblet cells in colon tissue was observed by AB-PAS staining.Results Compared with the normal group,the levels of serum ALP,ALT,AST,TBil,and DBil,the relative expression of IL-6 mRNA in liver tissue,and the average optical density value of TGR5 positive expression in colon tissue in the model group were significantly increased(all P<0.05),the relative expressions of cAMP and PKA mRNA in liver tissue and the average optical density value of TGR5 positive expression in liver tissue were significantly decreased(all P<0.05),and the number of
