摘要 目的探讨血液保存前不同温度及暴露时间对血液质量的影响。方法选择2020年5月至2022年3月赣州市中心血站血液样本236份作为研究对象,根据血液保存前温度不同分为2℃组和6℃组;根据血液保存前暴露时间不同分为暴露2h组和暴露4h组。分别于采血后第0、4、8、16天采用薄层色谱分析法检测膜磷脂水平;采用同步法检测红细胞膜Na^(+)-K^(+)-ATP酶;采用纯化心肌环腺苷酸磷二酯酶(PDE)法检测红细胞浆钙调素(CaM);分光光度法测定红细胞膜过氧化物(LPO)水平等。根据血液质量分为良好组与一般组,查阅血液标本不同参数,对可能的影响因素进行单因素和多因素Logistic回归分析。结果2℃组在各时间检测点的游离Hb(FHb)、K^(+)、LPO水平均低于6℃组(均P<0.05);pH、膜磷脂、CaM及Na^(+)-K^(+)-ATP酶水平均高于6℃组(均P<0.05);暴露2 h组在各时间检测点的FHb、K^(+)、LPO水平均低于暴露4h组(均P<0.05);pH、膜磷脂、CaM及Na^(+)-K^(+)-ATP酶水平均高于暴露4 h组(均P<0.05);单因素及多因素Logistic回归分析表明血液转运、血液保存前温度、血液保存前暴露时间、血液采集专业技能均为血液保存质量的独立影响因素(均P<0.05)。结论血液保存前不同温度及暴露时间会对血液质量产生影响,低温、暴露时间短更有利于保存血液质量。此外,血液转运、血液采集技术也会影响血液质量。 Objective To investigate the influence of different temperature and exposure time before blood preservation on blood quality.Methods A total of 236 blood samples were selected from May 2020 to March 2022 in Ganzhou City Central Blood Station as the research objects,which were divided into 2℃1 and 6℃groups according to the different temperatures before blood preservation,and into 2 h and 4 h groups according to the different exposure times before blood preservation.The levels of membrane phospholipids were measured by thin-layer chromatography on the 0th,4th,8th and 16th days after blood collection;the erythrocyte membrane Na^(+)-K^(+)-ATPase was measured by simultaneous assay;erythrocyte plasma calmodulin(CaM)was measured by the purified cardiac cyclic adenosine phosphodiesterase(PDE)method;and the levels of erythrocyte membrane peroxides(LPO)were determined by spectrophotometry,etc.The blood samples were divided into good and average groups according to the quality of the blood.According to the blood quality was divided into good and average groups,different parameters of the blood specimens were consulted,and the possible influencing factors were analyzed by univariate and multivariate logistic regression.Results The levels of free Hb(FHb),K^(+),and LPO in the 2℃group were lower than those in the 6℃group at all time detection points(all P<0.05);the levels of pH,membrane phospholipids,CaM,and Na^(+)-K^(+)-ATPase were higher than those in the 6℃group(all P<0.05);and the levels of FHb,K^(+),and LPO in the 2 h exposure group were lower than those in the 4 h exposure group at all time detection points(all P<0.05);The levels of pH,membrane phospholipids,CaM and Na^(+)-K^(+)-ATPase were higher than those in the 4-h exposure group(all P<0.05);single-factor and multifactorial logistic regression analysis showed that blood transfer,blood preservation temperature,blood exposure time,and professional skills in blood collection were all independent influences on the quality of blood preservation(all P<0.05).Conclu
