摘要
目的:分析枸杞多糖对绝经后骨质疏松患者肠道菌群结构和短链脂肪酸的影响。方法:通过人体肠道微生物系统模拟装置(SHIME■),构建Triple-SHIME■模拟近端结肠和远端结肠,分别接种3名绝经后骨质疏松症患者粪便样本,待稳定期后添加枸杞多糖(10g/L)连续干预2周。分别于稳定期第2周(Sw2)、治疗期的第1周和第2周(Tw1,Tw2)收集反应器中发酵液进行16S rRNA基因测序和序列分析,分析菌群结构。利用液相色谱串联质谱检测体外发酵液中短链脂肪酸的浓度变化。结果:与稳定期第2周取样结果相比,枸杞多糖干预后的发酵液放线菌门的相对丰度呈降低趋势,变形菌门的相对丰度呈上升趋势。乳杆菌属显著增殖(P<0.05),尤其在近端结肠中富集。总短链脂肪酸、乙酸、丙酸、和丁酸水平在枸杞多糖干预后显著高于稳定期(P<0.05)。结论:枸杞多糖可能通过调整肠道菌群结构,促进绝经后骨质疏松患者短链脂肪酸的生成,为临床进一步研究提供理论依据。
Objective:To investigate the effects of Lycium barbarum polysaccharide(LBP)on the intestinal flora structure and short-chain fatty acids in postmenopausal osteoporosis patients.Method:Triple-SHIME■simulated proximal colon and distal colon were constructed by simulator of human intestinal microbial ecosystem(SHIME■),inoculated with fecal samples from 3 PMO patients respectively,and then supplemented with LBPs(10g/L)for 2 weeks after the stable period.16S rRNA gene sequencing and sequence analysis were performed on the fermentation broth collected from the reactor at the 2nd week of the stable stage(Sw2)and the 1st and 2nd week of the treatment stage(Tw1,Tw2),respectively.The concentration of short-chain fatty acids in fermentation broth in vitro was determined by liquid chromatography-tandem mass spectrometry.Results:Compared with the sampling results at the 2nd week of stable period,the relative abundance of Actinobacteria tended to decreased and that of Proteobacteria increased.Lactobacillus spp.proliferated significantly,especially enriched in the proximal colon.The levels of total short-chain fatty acids,acetic acid,propionic acid and butyric acid were significantly higher after the intervention of LBPs(P<0.05).Conclusion:LBP promote the production of short-chain fatty acids in postmenopausal osteoporosis patients by adjusting the structure of intestinal flora.These results provide a theoretical basis for further clinical research.
