短期饥饿胁迫对摄入红枣提取物的虹鳟非特异性免疫相关基因表达的影响

摘要 为了解红枣提取物对虹鳟非特异性免疫能力影响的持续性和耐饥饿胁迫能力的影响,分别用含有低(0.25%)、高(0.50%)浓度红枣提取物的饲料和空白对照饲料连续饲喂虹鳟8周后禁食2周,然后对虹鳟脾脏、头肾的非特异性免疫相关基因表达进行检测,发现与对照组相比,在所测定的七个基因中,红枣提取物饲喂后的虹鳟经短期饥饿胁迫后C3、 GPX1、 GSTA和LZM四个基因存在显著差异,SOD、 CAT和TNF-α基因无显著变化。其中脾脏和头肾中C3基因在低浓度下极显著上调,而高浓度组脾脏C3基因表达量显著下降;脾脏中GPX1、 GSTA基因表达在高浓度组也均显著下调,仅LZM基因表达显著上调;而低浓度组这四个基因表达量均表现为一定程度的上调。结果表明较低浓度(0.25%)红枣提取物摄入对虹鳟短期饥饿胁迫有一定的非特异性免疫增强效果。 The experiment was conducted to study the effects of jujube extracts on the persistence of nonspecific immunity in rainbow trout and its tolerance to starvation stress.Rainbow trout were continuously fed with feed containing low(0.25%)and high(0.50%)concentration of jujube extracts and blank control for 8 weeks,then fasted for 2 weeks.After that the expression of nonspecific immune related gene in rainbow trout spleen and head kidney were detected.The results relative to blank control groups showed that C3,GPX1,GSTA,LZM had significant differences;and SOD,CAT,TNF-α had no significant differences.The immune related gene C3 in spleen and head kidney were up-regulated very significantly in low(0.25%)groups,but which in spleen down-regulated significantly in high(0.50%)groups.The gene expression of GPX1,GSTA in spleen were down-regulated significantly too but LZM in spleen were up-regulated significantly in high(0.50%)groups.And all of the four gene expression were up-regulated to a certain extent in low(0.25%)groups.The findings indicated that intake of lower jujube extract(0.25%)had a positive effect to some extent on nonspecific immunity in rainbow trout which were dealt with short-term starvation stress.
