紫锥菊提取物菊苣酸对低氧环境下H9c2大鼠心肌细胞增殖的影响

Effects of echinacea purpurea extract on proliferation of H9c2 rat cardiomyocytes under hypoxic environment

摘要 探究低氧条件下,菊苣酸(cichoric acid,CA)对H9c2大鼠心肌细胞增殖的影响。构建体外H9c2大鼠心肌细胞低氧模型,设常氧组、低氧组、低氧+CA组和低氧+红景天苷组,流式细胞术检测H9c2大鼠心肌细胞的细胞周期,qRT-PCR和Western blot检测增殖相关因子周期蛋白T2(cyclin T2,CCNT2)、周期蛋白依赖性激酶9(cyclin-dependent kinases 9,CDK9)和叉头盒蛋白1(forkhead box prote1,FOXP1)的mRNA和蛋白表达。结果显示,与常氧组相比,低氧组G0/G1期细胞数显著增加(P<0.05),S期和G2/M期细胞数显著降低(P<0.05),极显著下调CCNT2、CDK9和FOXP1的mRNA和蛋白表达(P<0.01)。与低氧组相比,低氧+CA组G0/G1期细胞数显著降低(P<0.05),G2/M期细胞数均显著增加(P<0.05),显著上调CCNT2(P<0.01)mRNA表达,上调CDK9和FOXP1的mRNA表达(P<0.01)和蛋白表达(P<0.05);低氧+红景天苷组G0/G1期细胞数显著降低(P<0.05),S期和G2/M期细胞数均显著增加(P<0.05),极显著上调CCNT2、CDK9和FOXP1的mRNA表达(P<0.01),显著上调CCNT2和CDK9的蛋白表达(P<0.05),极显著上调FOXP1的蛋白表达(P<0.01)。结果表明,CA能通过促进增殖相关因子CCNT2、CDK9和FOXP1的表达,有效保护低氧诱导的H9c2大鼠心肌细胞增殖活性,缓解低氧诱导的H9c2大鼠心肌细胞周期阻滞现象,为CA作为低氧保护剂和畜牧业新型植物源低氧保护性饲料添加剂的开发利用提供理论参考。 To investigate the effect of cichoric acid(CA)on the proliferation of H9c2 rat cardiomyocytes under hypoxia.The cells were divided into the normoxia group,hypoxia group,hypoxia+CA group,and the hypoxia+salidroside group,the mRNA and protein expressions of Cyclin T2,CDK9 and Foxp1 were detected by qRT-PCR and Western blot.Compared with the normoxia group,the number of cells in G0/G1 phase was significantly increased(P<0.05),and the number of cells in S phase and G2/M phase was significantly decreased(P<0.05),the mRNA and protein expressions of CCNT2,CDK9 and FOXP1 were significantly down-regulated(P<0.01).Compared with the hypoxia group,the cells in G0/G1 phase decreased significantly(P<0.05),and the cells in G2/M phase increased significantly(P<0.05)in the hypoxia+CA group,and the expression of CCNT2 mRNA was upregulated significantly(P<0.01),the mRNA and protein expressions of CDK9(P<0.01)and FOXP1(P<0.05)were up-regulated,the cell numbers in G0/G1 phase were significantly decreased(P<0.05),and the cell numbers in S phase and G2/M phase were significantly increased(P<0.05)in hypoxia+salidroside group,significantly upregulated the mRNA expression of FOXP1(P<0.01),the protein expression of CCNT2and CDK9(P<0.05),and the protein expression of FOXP1(P<0.01).CA can effectively protect the proliferation of H9c2 rat cardiomyocytes induced by hypoxia by promoting the expression of proliferation-related factors CCNT2,CDK9and FOXP1,to alleviate the hypoxia-induced cycle arrest of H9c2 rat cardiomyocytes,and to provide a theoretical reference for the development and utilization of CA as a hypoxic protective agent and a new type of plant-derived hypoxic protective feed additive for animal husbandry.
