摘要
目的研究暗褐网柄牛肝菌Phlebopus portentosus的化学成分及其抗肿瘤细胞毒活性。方法暗褐网柄牛肝菌95%乙醇提取物采用硅胶、ODS、半制备HPLC行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法测定其对人癌细胞株HepG2、A549、HeLa、MCF-7、MDA-MB-231的细胞毒活性。结果从中分离得到4个化合物,分别鉴定为phlesterol A(1)、过氧化麦角甾醇(2)、(22 E,24R)-7,22-二烯-3β,5α,6β-麦角三醇(3)、麦角甾-7,22 E-二烯-3-酮(4)。化合物1对HeLa、MCF-7细胞的抑制活性较强,IC 50值为6.45~8.38μmol/L,对MDA-MB-231细胞具有一定的细胞毒活性,IC 50值为14.84μmol/L。结论化合物1为新化合物,其对妇科肿瘤细胞有一定的抑制活性。
AIM To study the chemical constituents from Phlebopus portentosus and their anti-tumor cytotoxic activities.METHODS The 95%ethanol extract from P.portentosus was isolated and purified by silica gel,ODS and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their cytotoxic activities of human cancer cell lines HepG2,A549,HeLa,MCF-7,MDA-MB-231 were evaluated by MTT assay.RESULTS Four compounds were isolated and identified as phlesterol A(1),ergosterol peroxide(2),(22 E,24R)-7,22-diene-3β,5α,6β-ergosta triol(3),ergosta-7,22 E-diene-3-one(4).Compound 1 had strong inhibitory activities on HeLa and MCF-7 cells,with IC 50 values ranging from 6.45 to 8.38μmol/L,and had certain cytotoxic activity against MDA-MB-231 cells,with an IC 50 value of 14.84μmol/L.CONCLUSION Compound 1 is a new compound,and displayes obvious inhibitory activity against gynecological cancer cells.
