摘要
目的为蒙药额尔敦-乌日勒的毒性及安全性研究提供参考。方法采用固定剂量法,将32只SD大鼠随机分为对照组(等体积纯净水)及额尔敦-乌日勒低、中、高剂量组(0.54,2.16,5.40 g/kg,简称低、中、高剂量组),各8只,分别灌胃相应药物或纯净水后观察14 d。给药前禁食12 h,不禁饮。观察大鼠一般情况与行为、饮食饮水、体质量及主要器官(心、肝、脾、肺、肾)情况(外观、脏器相对质量、组织形态学变化等),检测血常规及血生化指标。结果各组大鼠用药后均未见中毒症状或死亡。各组大鼠体质量和主要器官相对质量均整体保持自然增长,且组间无明显差异(P>0.05),但各组雌雄大鼠间存在不同程度差异。体质量变化率,用药第7,14天,低剂量组雄性大鼠明显低于对照组(P<0.05),中剂量组雌性大鼠明显高于对照组(P<0.01)。各组大鼠各主要器官颜色、形态无明显差异;中、高剂量组雄性大鼠肾脏相对质量及高剂量组雌性大鼠脾脏相对质量均明显低于对照组(P<0.05)。各组大鼠主要器官病理形态学无明显异常。中、高剂量组雄性大鼠血常规及血及生化指标仅粒细胞计数及白蛋白含量与对照组有明显差异(P<0.05),但均无生物学意义。结论经初步研究,大鼠对额尔敦-乌日勒单次给药的最高耐受量可达5.40 g/kg。
Objective To provide a reference for the toxicity and safety study of Mongolian medicine Eerdun-Wurile.Methods Thirty-two SD rats were randomly divided into the control group(equal volume of purified water)and the Eerdun-Wurile low-,medium-and high-dose groups(0.54,2.16,5.40 g/kg,hereinafter referred to as the low-,medium-and high-dose groups)by the fixed dose procedure,with eight rats in each group.They were given corresponding drugs or purified water by gavage and observed for 14 d.All rats were fasted for 12 h before the administration,but were not prohibited from drinking.The general condition,behavior,diet,drinking,body mass,and major organs(heart,liver,spleen,lung,kidney)condition(such as appearance,relative mass of organs,histomorphological changes)of rats were observed.The blood routine indicators and biochemical indicators in rats were detected.Results After medication,no toxic symptom or death was observed in rats.The body mass and relative mass of major organs in each group maintained a natural increase overally,with no significant difference between groups(P>0.05),while there were differences between female and male rats in each group.The body mass change rate of male rats in the low-dose group was significantly lower than that of male rats in the control group on the 7th and 14th days after medication(P<0.05),while the above indicator of female rats in the medium-dose group was significantly higher than that of female rats in the control group on the 7th and 14th days after medication(P<0.01).There was no significant difference in the color and morphology of the major organs in each group.The kidney relative mass of male rats in the medium-and high-dose groups and the spleen relative mass of female rats in the high-dose group were significantly lower than those in the control group(P<0.05).There were no significant abnormality in the pathological morphology of the major organs in each group.In blood routine indicators and biochemical indicators,only granulocyte count and albumin content of male rats
