摘要
采用网络药理学、生物信息学并结合动物试验的方法,探讨七味白术散治疗脾虚泄泻小鼠的潜在作用机制。通过TCMSP数据库筛选和预测七味白术散的活性成分及潜在靶点,应用GEO数据库中的GSE14841数据集、GeneCards、OMIM和DisGeNET数据库收集脾虚泄泻相关疾病靶点,利用韦恩图获取药物和疾病的交集靶点,通过Cytoscape软件构建交集靶点的PPI网络,采用Metascape数据库对交集靶点进行GO和KEGG富集分析,最后通过脾虚泄泻小鼠模型进行验证。结果:七味白术散筛选得到149个活性成分,共有367个靶点;脾虚泄泻共有742个潜在的治疗靶点,两者共有36个交集靶点;GO和KEGG分析显示交集靶点主要通过调控细胞凋亡、T细胞受体信号通路、炎症性肠病和TNF等信号通路改善脾虚泄泻,主要涉及炎症反应、免疫调节等相关生物进程;动物试验发现七味白术散可明显改善脾虚泄泻小鼠的腹泻症状,增加其体重,减轻其结肠组织的病理损伤,并上调了小鼠结肠屏障蛋白Claudin-1、Occludin和血清中D-木糖的表达,证实了整合分析的可靠性。本试验表明,七味白术散可通过“多成分,多靶点,多途径”发挥治疗脾虚泄泻的作用,为阐释七味白术散治疗脾虚泄泻的临床应用提供了科学依据。
Using network pharmacology combined with bioinformatics and with animal experiments,the potential mechanism of Qiwei Baizhu Powder in the treatment of spleen-deficiency diarrhea(SDD)mice was explored in this study.The active ingredients and potential targets of Qiwei Baizhu Powder were screened and predicted through the TCMSP database.The GSE14841 data set in the GEO database,GeneCards,OMIM and DisGeNET databases were used to collect disease targets related to SDD.The Venn diagram was used to get the intersection targets,and the PPI network of the intersection targets was constructed through Cytoscape software.The Metascape database was used to conduct GO and KEGG enrichment analyses on the intersection targets,and finally SDD mice models were used for verification.The results were as follows:149 active ingredients were screened out from Qiwei Baizhu Powder,with a total of 367 targets.There are 742 potential therapeutic targets for SDD,and 36 overlapping targets between the two.The GO and KEGG analyses showed that the intersection targets improved SDD mainly by regulating apoptosis,the T cell receptor signaling pathway,TNF and other signaling pathways in the inflammatory bowel disease,and by involving related biological processes,such as inflammatory response and immune regulation.The animal experiments found that Qiwei Baizhu Powder significantly improved the diarrhea symptoms of the SDD mice,increased their body weight,reduced the pathological damage of their colon tissue,and up-regulated the expression of mouse colonic barrier proteins Claudin-1,Occludin and D-xylose in their serum.Qiwei Baizhu Powder played a role in treating SDD through“multiple components,multiple targets,and multiple channels”.This study provided a scientific support for interpreting clinical research on Qiwei Baizhu Powder in treating SDD.
