摘要
动脉粥样硬化是心肌梗死、脑卒中等急性心脑血管事件的主要原因,是多种细胞参与的复杂病理过程。动脉血管平滑肌细胞(vascular smooth muscle cells, VSMCs)的增殖、迁移与表型转化是形成动脉粥样硬化斑块的核心机制之一。近些年,细胞谱系追踪和单细胞RNA测序等新技术的出现几乎颠覆了以往对VSMC在斑块中表型和功能的理论认知。本文将系统阐述动脉粥样硬化疾病进程中VSMC的表型转化及其调控机制,新兴技术对该领域的推动作用,以期进一步探索VSMC在动脉粥样硬化中的作用。
Atherosclerosis is the main cause of acute cardiovascular and cerebrovascular events such as myocardial infarction and stroke.It is a complex pathological process involving many kinds of cells.The proliferation,migration and phenotypic switching of vascular smooth muscle cells(VSMCs)are one of the important mechanisms of atherosclerotic plaque formation.In recent years,new technologies such as cell lineage tracing and single-cell RNA sequencing have almost overturned our previous knowledge of the function of VSMCs in plaques.In this review,we will systematically describe VSMCs phenotypic switching and its regulatory mechanism in the process of atherogenesis,and introduce emerging technologies in this field,so as to further explore the role of VSMCs in atherosclerosis.
