摘要
利用固定化脂肪酶对玉米毛油进行酶解,采用单因素试验和正交试验研究了酶解温度、酶添加量、水添加量、甘油添加量以及酶解时间对1,3-甘油二酯含量的影响,同时测定了分子蒸馏过程中二级蒸馏温度、进料流速以及刮膜转速对分离纯化1,3-甘油二酯的影响。结果表明:固定化脂肪酶酶解玉米毛油的最优酶解条件为酶解温度70℃、酶添加量0.3%、水添加量20%、甘油添加量10%以及酶解时间4 h。在进行分子蒸馏时,在冷凝温度,真空度一定的条件下,确定了二级分子蒸馏的最佳分离纯化条件,即蒸馏温度200℃、进料流速5.5 mL/min和刮膜转速300 r/min,在此条件下,能高效提高1,3-甘油二酯的含量,提高产品的品质。
Corn crude oil was enzymatically hydrolyzed by immobilized lipase.The effects of enzymolysis temperature,enzyme addition amount,water addition amount,glycerin addition amount,and enzymolysis time on the content of 1,3-diglyceride were studied by single factor experiment and orthogonal experiments.At the same time,the effects of secondary distillation temperature,material flow rate,and scraper speed on the separation and purification of 1,3-diglyceride were measured.The results showed that the optimal enzymatic hydrolysis conditions for corn crude oil using immobilized lipase were as follows:enzymatic hydrolysis temperature 70℃,enzyme addition 0.3%,water addition 20%,glycerol addition 10%,and enzymatic hydrolysis time 4 hours.Under the conditions of condensation temperature and vacuum degree,the best separation and purification conditions of secondary molecular distillation were a distillation temperature of 200℃,a material flow rate of 5.5 mL/min,and a scraping speed of 300 r/min.Under these conditions,the content of 1,3-diglycerid could be effectively improved and the quality of products can be improved.
