摘要
目的探讨巢蛋白(nestin)在胃癌组织中的表达及其与患者预后的关系。方法采用免疫组织化学染色检测137例胃癌组织和20例癌旁正常组织中nestin蛋白表达,定量PCR检测12例胃癌组织和癌旁正常组织中nestin mRNA表达,分析nestin表达与胃癌患者临床病理特征的关系。采用log-rank检验分析胃癌患者总生存期,Cox风险比例模型分析胃癌患者预后的影响因素。结果胃癌组织nestin阳性表达率和mRNA表达高于癌旁正常组织(P<0.01)。Nestin阳性表达与病理分级、浸润深度、淋巴结转移、远处转移和TNM分期有关(P<0.05)。Nestin阳性表达患者总生存期为(55.46±5.23)个月,短于nestin阴性表达患者的(93.51±5.15)个月(P<0.01)。远处转移,TNM分期Ⅲ、Ⅳ期及nestin阳性表达是胃癌患者死亡的独立危险因素(P<0.05)。结论Nestin的表达与胃癌患者浸润深度、转移和分期有关,可用于评估胃癌患者预后。
Objective To investigate the expression of nestin in gastric cancer tissues and its relationship with the prognosis of the patients with gastric cancer.Methods The protein expression of nestin was detected in 137 gastric cancer tissues and 20 tumor-adjacent normal tissues by immunohistochemistry.The mRNA expression of nestin in 12 gastric cancer tissues and tumor-adjacent normal tissues were detected by quantitative PCR.The relationship between nestin protein expression and the clinicopathologic features of the patients with gastric cancer was analyzed.The overall survival of the gastric cancer patients was analyzed by log-rank test.The influencing factors for the prognosis of the patients with gastric cancer were analyzed by Cox risk proportional model.ResultsThe mRNA expression and positive expression rate of nestin in gastric cancer tissues were higher than those in tumor-adjacent normal tissues(P<0.01).The positive expression of nestin was correlated with pathological grade,depth of invasion,lymph node metastasis,distant metastasis and TNM stage(P<0.05).The overall survival of the patients with nestin positive expression was(55.46±5.23)months,which was shorter than that of(93.51±5.15)months in the patients with nestin negative expression(P<0.01).Distant metastasis,TNM stageⅢandⅣand nestin positive expression were the independent risk factors for death in the patients with gastric cancer(P<0.05).Conclusion The expression of nestin is related to the depth of invasion,metastasis and staging in gastric cancer,and can be used to evaluate the prognosis of the patients with gastric cancer.
