摘要
目的探讨高尿酸血症(HUA)对雌性大鼠生殖功能的影响。方法利用腺嘌呤、高酵母饲料和氧嗪酸钾(尿酸酶抑制剂)进行9个周的HUA模型建立。通过阴道脱落细胞涂片观察大鼠动情周期;留取完整子宫、卵巢称质量,计算器官系数;采用苏木精-伊红(HE)染色观察卵巢的病理变化;采用实时荧光定量PCR(qPCR)方法检测卵巢组织中吻素1(KISS1)和吻素1受体(KISS1R)mRNA表达。结果HUA雌性大鼠模型构建成功。与正常大鼠相比,HUA雌性大鼠动情间期明显缩短(F=14.000,P<0.01),卵巢系数显著下降(t=2.377,P<0.05),卵巢组织中KISS1和KISS1R mRNA表达显著增高(t=3.456、5.408,P<0.05)。结论HUA可造成雌性大鼠KISS1系统紊乱,引起动情间期缩短和卵巢系数降低。
Objective To investigate the effects of hyperuricemia(HUA)on reproductive function in female rats.[WTHX]Methods A HUA model was established by nine weeks of high-yeast feeding with adenine and oteracil potassium(a uricase inhibitor).The rat estrous cycle was determined through observing the smears of exfoliated vaginal cells.The intact uterus and ovaries were weighed to calculate the organ coefficients.Pathological changes in the ovaries were observed with hematoxylin-eosin(HE)staining.The mRNA expression of kisspeptin 1(KISS1)and kisspeptin 1 receptor(KISS1R)in the ovaries was measured using quantitative real-time PCR.Results The female rat HUA model was successfully constructed.Compared with normal rats,the female rats with HUA showed a significantly shorter interestrus interval(F=14.000,P<0.01),a significantly lower ovarian coefficient(t=2.377,P<0.05),and significantly higher mRNA expression of KISS1(t=3.456,P<0.05)and KISS1R(t=5.408,P<0.05)in the ovarian tissue.Conclusion HUA can disrupt the KISS1 system in female rats,resulting in a shortened interestrus interval and a reduced ovarian coefficient.
