31~60日龄中速型黄羽肉鸡异亮氨酸需要量研究 被引量:1

Study on Isoleucine Requirement of Medium⁃Growing Yellow⁃Feathered Chickens Aged from 31 to 60 Days

摘要 本试验旨在研究饲粮不同异亮氨酸水平对31~60日龄中速型黄羽肉鸡生长性能、胴体性状、血浆生化指标、肠道氨基酸转运以及蛋白质代谢和肝脏脂质代谢相关基因表达的影响,并确定其适宜需要量。试验选用1080只初始体重为(649.17±0.84)g的31日龄黄羽肉鸡母鸡,根据体重一致原则随机分为6个组,每组6个重复,每个重复30只鸡。各组饲粮异亮氨酸水平分别为0.58%、0.66%、0.74%、0.82%、0.90%和0.98%。试验期30 d。结果表明:1)随着饲粮异亮氨酸水平的升高,黄羽肉鸡料重比和屠宰率呈线性和二次变化(P<0.05),腹脂率呈线性降低(P<0.05),半净膛率和全净膛率显著升高(P<0.05)。2)随着饲粮异亮氨酸水平的升高,黄羽肉鸡血浆白蛋白(ALB)和总胆固醇(TC)含量呈线性和二次变化(P<0.05),血浆尿酸(UA)含量显著降低(P<0.05)。3)随着饲粮异亮氨酸水平的升高,黄羽肉鸡空肠和回肠小肽转运载体1(PepT1)mRNA相对表达量呈线性和二次变化(P<0.05),回肠20S蛋白酶体(20SP)mRNA相对表达量显著降低(P<0.05)。4)随着饲粮异亮氨酸水平的升高,黄羽肉鸡肝脏3-羟基3-甲基戊二酰辅酶A还原酶(HMGCR)mRNA相对表达量呈线性和二次变化(P<0.05),肝脏肉碱棕榈酰转移酶1(CPT1)mRNA相对表达量呈二次变化(P<0.05),肝脏沉默信息调节因子1(SIRT1)mRNA相对表达量显著升高(P<0.05)。综上所述,饲粮异亮氨酸水平显著影响黄羽肉鸡母鸡生长性能和胴体性状,且适宜水平的异亮氨酸可上调肠道氨基酸转运和肝脏脂肪分解相关基因表达,抑制蛋白质分解和脂肪生成相关基因表达。依据二次曲线回归模型估测,31~60日龄中速型黄羽肉母鸡获得最佳生长性能和胴体性状的饲粮异亮氨酸适宜水平为0.76%~0.83%;其中,以料重比为主要评价指标,黄羽肉鸡母鸡饲粮异亮氨酸适宜水平推荐为0.76%,每日需要量为735 mg。 This experiment was conducted to investigate the effects of different dietary isoleucine levels on growth performance,carcass traits,plasma biochemical indices,intestinal amino acid transport and protein me-tabolism as well as liver lipid metabolism related gene expression of medium-growing yellow-feathered chickens aged from 31 to 60 days,and to determine the appropriate requirement.A total of 1080 female yellow-feath-ered chickens of 31-day-old with an initial body weight of(649.17±0.84)g were randomly divided into 6 groups with 6 replicates per group and 30 chickens per replicate.The dietary isoleucine levels were 0.58%,0.66%,0.74%,0.82%,0.90%and 0.98%,respectively.The experiment lasted for 30 days.The results showed as follows:1)with dietary isoleucine level increasing,the ratio of feed to gain and dressing rate of yellow-feathered chickens had linear and quadratic changes(P<0.05),the abdominal fat rate was linearly de-creased(P<0.05),and the rates of half-eviscerated and eviscerated were significantly increased(P<0.05).2)With the dietary isoleucine level increasing,the plasma contents of albumin(ALB)and total cholesterol(TC)of yellow-feathered chickens had linear and quadratic changes(P<0.05),and the plasma uric acid(UA)content was significantly decreased(P<0.05).3)With dietary isoleucine level increasing,the small peptide transporter 1(PepT1)mRNA relative expression level in jejunum and ileum of yellow-feathered chick-ens had linear and quadratic changes(P<0.05),and the 20S proteasome(20SP)mRNA relative expression level in ileum was significantly decreased(P<0.05).4)With dietary isoleucine level increasing,the 3-hy-droxy-3-methylglutaryl-CoA reductase(HMGCR)mRNA relative expression level in liver of yellow-feathered chickens had linear and quadratic changes(P<0.05),the carnitine palmitoyl transterase 1(CPT1)mRNA rel-ative expression level in liver had quadratic changes(P<0.05),and the silencing information regulator 1(SIRT1)mRNA relative expression level in liver was significantly increased(P<0.05).In conc
