深色有隔内生真菌对干旱胁迫下樟子松生长及抗旱性的影响

摘要 【目的】探究干旱胁迫下深色有隔内生真菌(Dark septate endophytes,DSE)对樟子松Pinus sylvestris var.mongolica幼苗的接种效应。【方法】采用棚内盆栽试验,以2年生樟子松幼苗为材料,进行J09、J32、J35、A065和灭菌PD培养液5种接种处理,设置正常浇水(WW)、中度干旱胁迫(MW)和重度干旱胁迫(LW)3种干旱处理,观测不同处理下DSE定殖形态,测定幼苗生长指标、幼苗生理指标、根际土壤理化性质及根际土壤酶活性。【结果】随着干旱胁迫的加剧,菌丝直径变粗,横隔变短,微菌核增多;与不接种相比,在中度干旱胁迫时,DSE对樟子松的促生作用最显著,樟子松的地径、地上鲜质量、地下鲜质量、地上干质量和地下干质量最高分别提高了67.72%、36.13%、41.58%、36.13%和41.48%;与不接种相比,干旱胁迫下,接种DSE显著提高樟子松幼苗超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)的活性和谷胱甘肽(GSH)的含量,最高分别可达229.60%、67.80%、28.16%和37.83%;接种DSE可显著降低幼苗丙二醛(MDA)、可溶性糖(PSS)和脯氨酸(PRO)的含量,与未接种相比,最高分别为74.16%、105.91%和51.24%;DSE能提高土壤有机质(OM)、全氮(TN)、速效氮(AN)、全磷(TP)、速效磷(AP)、全钾(TK)和速效钾(AK)的含量,与未接种相比,最高分别增加了19.85%、40.64%、35.25%、57.92%、48.11%、55.62%和68.84%;与未接种相比,DSE还能不同程度地提高土壤过氧化氢酶(S-CAT)、蔗糖酶(S-SC)和脲酶(S-UE)活性,最高分别可达71.85%、128.64%和65.05%,特别是干旱胁迫下,接种J09和A065对寄主植物的生物量、形态和生理指标有显著的正向影响;接种J09和A065后,根际土壤酶活性和理化性质显著提高,而接种J35显著抑制了樟子松幼苗的生长指标和生理指标。【结论】DSE定殖形态的变化可能是提高寄主植物的耐旱机制之一,DSE的定殖种类显著影响寄主植物的耐旱性;接种J09和A065可以通过改 【Objective】The objective of this study was to understand the colonization effects of dark septate endophytes on Pinus sylvestris var.mongolica under drought stress.【Method】With the pot experiment in the greenhouse,the 2-year-old P.sylvestris var.mongolica seedlings were inoculated with J09,J32,J35,A065 and the sterile PD medium.Under normal watering(WW),moderate drought(MW)and severe drought(LW)conditions,the difference in the colonization morphology of DSE,the growth index of seedlings,physiological indexes of seedlings,physical and chemical properties of rhizosphere soil,and enzyme activities of rhizosphere soils were studied.【Result】With the intensification of drought stress,the diameter of hyphae became thicker,the septum became shorter,and the number of microsclerotia increased.The inoculation of DSE enhanced the ground diameter,fresh weight of the aboveground part,fresh weight of the underground part,dry weight of the aboveground part and dry weight of the underground part of P.sylvestris var.mongolica by 67.72%,36.13%,41.58%,36.13%,and 41.48%under the MW treatments compared with CK.The inoculation of DSE promoted the CAT,POD,SOD and GSH activities of the plant by 229.60%,67.80%,28.16%and 29.92%,respectively,under drought stress,in comparison with CK.The inoculation of DSE elevated the content of MDA,PSS and PRO of the P.sylvestris var.mongolica seedings by 74.16%,105.91%,and 51.24%,respectively,in comparison with CK.DSE increased the contents of the soil OM,TN,AN,TP,AP,TK and AK by 19.85%,40.64%,35.25%,57.92%,48.11%,55.62%,and 68.84%,respectively,in comparison with CK.DSE also increased the S-CAT,S-SC,and S-UE activities by 71.85%,128.64%,and 65.05%,respectively,compared with CK.Specifically,the inoculation of J09 and A065 significantly increased the growth and physiological indexes of seedlings,the physical and chemical properties of rhizosphere soils,and the enzyme activity of rhizosphere soils under drought stress.However,the inoculation of J35 significantly inhibited the growth and physiolo
