产AmpC酶的猪源奇异变形杆菌分离鉴定及生物学特性分析 被引量:3

摘要 奇异变形杆菌(Proteus mirabilis)是一种可导致多种动物感染的条件致病菌,目前有关产AmpC酶的动物源奇异变形杆菌研究较少。为探究广西某猪场母猪流产、死胎的原因,从流产猪胎肝脏中分离到病原菌,对分离株进行纯化培养、染色镜检、生化试验和16S rRNA测序确定为奇异变形杆菌,命名为GX-Y9251株。对GX-Y9251株进行药敏试验、耐药基因检测、AmpC酶基因检测、毒力基因检测、小鼠致病性试验和生物膜半定量检测。药敏试验结果显示,该菌株对8种药物敏感,对17种抗生素耐药。GX-Y9251株存在多种耐药基因,且产AmpC酶,为blaDHA基因。GX-Y9251株存在6个毒力基因,其对小鼠的半数致死量(LD50)为5.4×107CFU;具有生物被膜形成能力,为中等黏附型(++)。研究结果为奇异变形杆菌感染的防治提供了理论依据。 Proteus mirabilis is an opportunistic pathogen that causes many kinds of animal infections.At present,there are few studies on animal derived Proteus mirabilis producing AmpC enzyme.In order to determine the causes of abortion and stillbirth of sows in a pig farm in the Guangxi area,pathogenic bacteria were isolated from the liver of an aborted pig.The isolated strains were purified and cultured,stained by microscopy,biochemical tests and 16S rRNA sequencing,and they were identified as Proteus mirabilis,named the GX-Y9251 strain.Then,we carried out a drug sensitivity test,drug resistance gene detection,AmpC enzyme gene detection,virulence gene detection,a mouse pathogenicity test and semi quantitative detection of biofilm for the GX-Y9251 strain.The results were that the drug sensitivity test showed the strain to be sensitive to 8 kinds of drugs,but resistant to the other 17 kinds of antibiotics.Many drug resistance genes were observe in the drug resistance detection,and the CX-Y9251 strain produced AmpC enzyme,the gene being blapHA.The strain had six virulence genes,and its LDso was 5.4×10^(7) CFU.It possessed the ability of biofilm formation which was of the medium adhesion type(++).The above results provide theoretical support for the prevention and treatment of Proteus mirabilis.
